Abstract

In this study, the complete mitogenome sequence of (Orthoptera: Tettigoniidae) has been decoded for the first time by next generation sequencing method. The overall base composition of mitogenome are 37.1% for A, 17.3% for C, 11.1% for G and 34.4% for T, and has high AT content of 71.5%. The complete mitochondrial genome of is 15,923 bp in length, and consisted of 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes and one A + T-rich region. An unusual feature of the mitogenome is the presence of a -like sequence on the J-strand, and the -like sequence have proper folding structures and anticodons sequences. Using the 13 PCGs and 2 rRNA of , together with 17 other close-related and two outgroup species, we constructed Bayesian inference phylogenetic tree to verify the accuracy and utility of new determined mitogenome sequences. The complete mitogenome of provides valuable molecular data for further phylogenetic and evolutionary analysis in Tettigoniidae.

在本研究中,首次通过下一代测序方法解码了(直翅目:螽斯科)的完整线粒体基因组序列。该线粒体基因组的总体碱基组成为:A占37.1%,C占17.3%,G占11.1%,T占34.4%,AT含量高达71.5%。该线粒体基因组全长15,923 bp,由13个蛋白质编码基因(PCGs)、22个tRNA基因、2个rRNA基因和1个富含A+T的区域组成。该线粒体基因组的一个不寻常特征是在J链上存在一个类似的序列,且该类似序列具有合适的折叠结构和反密码子序列。利用该物种的13个PCGs和2个rRNA,以及其他17个近缘物种和2个外类群物种,我们构建了贝叶斯推断系统发育树,以验证新测定的线粒体基因组序列的准确性和实用性。该物种的完整线粒体基因组为螽斯科进一步的系统发育和进化分析提供了有价值的分子数据。
