To study the Periplaneta americana L. extract Ento-B on the treatment of chronic ulcerative colitis induced by 2,4-dinitrochlorobenzene and acetic acid in rats and to explore its primary mechanism of action.
Using 2,4-dinitrochlorobenzene combined with acetic acid to induce chronic ulcerative colitis (chronic UC) in rats. The sulfasalazine (400 mg/kg) and Ento-B (200 mg/kg, 100 mg/kg,50 mg/kg) were given by intragastric administration and the effect was evaluated according to the disease activity index (DAI) score, colon mucosal injury index (CMDI) score, histopathological score (HS) and the serum levels of Interleukin-4(IL-4), Interleukin-10(IL-10), Tumor necrosis factor-α(TNF-α), Malondialdehyde(MDA), Superoxide dismutase(SOD) and Inducible nitric oxide synthase(iNOS.).
Compared with the model group, all doses of Ento-B could reduce the score of CMDI (p < 0.05), HS(p < 0.05 or p < 0.01), significantly increased the expression of IL-4, IL-10, SOD (p < 0.01) and decreased the levels of TNF-α, MDA, iNOS in serum of UC rats, significantly improving the degree of colon lesionsin UC rats.
Ento-B may play an important role in the treatment of ulcerative colitis induced byUC rats. The mechanism may be related to the increased expression of IL-4, IL-10, SOD and reduced expression of TNF-α, MDA, iNOS.
研究美洲大蠊提取物Ento-B对2,4-二硝基氯苯和乙酸诱导的大鼠慢性溃疡性结肠炎的治疗作用,并探讨其主要作用机制。
采用2,4-二硝基氯苯联合乙酸诱导大鼠慢性溃疡性结肠炎(慢性UC)。通过灌胃给予柳氮磺胺吡啶(400mg/kg)和Ento-B(200mg/kg、100mg/kg、50mg/kg),根据疾病活动指数(DAI)评分、结肠黏膜损伤指数(CMDI)评分、组织病理学评分(HS)以及血清白细胞介素-4(IL-4)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)、丙二醛(MDA)、超氧化物歧化酶(SOD)和诱导型一氧化氮合酶(iNOS)水平评估疗效。
与模型组相比,各剂量的Ento-B均可降低CMDI评分(p<0.05)、HS评分(p<0.05或p<0.01),显著增加UC大鼠血清中IL-4、IL-10、SOD的表达(p<0.01),降低TNF-α、MDA、iNOS水平,明显改善UC大鼠结肠病变程度。
Ento-B对UC大鼠诱导的溃疡性结肠炎可能具有重要治疗作用。其机制可能与IL-4、IL-10、SOD表达增加及TNF-α、MDA、iNOS表达降低有关。
