spp. have many applications in complementary medicine and are recognized as the most important sources of natural products for bone health and regeneration especially in postmenopausal women. Therefore, the aim of this study was to investigate the effects of the extracts from three species on proliferation and osteogenesis potential of human menstrual blood-derived mesenchymal stem cells (MenSCs).
The possible cytotoxic activity of three members of spp. (at concentrations of 5 to 100 μg/ml) was determined using MTT (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide) assay. Alkaline phosphatase (ALP) activity assay, Alizarin Red-S staining, and the expression analysis of an osteoblastic gene were performed to evaluate osteogenic differentiation potential.
The extracts of and decreased the viability and growth of MenSCs while increased the proliferation of cells after 72 hr incubation. Treatment of MenSCs with selected plant extracts revealed that and could enhance the osteogenic potential of MenSCs in terms of ALP activity. The expression in the presence of was significantly greater than observed following treatment with 17β-estradiol (as positive control).
The results of this study suggest that and may have therapeutic values as a nutraceutical with respect to their considerable influence on osteogenic potential of mesenchymal stem cells.
某属植物在补充替代医学中有多种应用,并且被认为是对骨骼健康和再生而言最重要的天然产物来源,尤其是对绝经后女性。因此,本研究的目的是调查三种该属植物提取物对人月经血来源间充质干细胞(MenSCs)增殖和成骨潜能的影响。
使用MTT(3-(4,5-二甲基噻唑-2-基)-2,5-二苯基四氮唑溴盐)法测定该属三种植物(浓度为5至100μg/ml)可能的细胞毒性活性。进行碱性磷酸酶(ALP)活性测定、茜素红S染色以及成骨细胞基因的表达分析以评估成骨分化潜能。
某两种植物提取物降低了MenSCs的活力和生长,而另一种植物提取物在孵育72小时后增加了细胞增殖。用选定的植物提取物处理MenSCs表明,就ALP活性而言,某两种植物提取物可增强MenSCs的成骨潜能。在该植物提取物存在的情况下,某基因的表达明显高于用17β-雌二醇(作为阳性对照)处理后的表达。
本研究结果表明,某两种植物提取物可能作为营养保健品具有治疗价值,因为它们对间充质干细胞的成骨潜能有相当大的影响。
