Extraction and Study of Hypoglycemic Constituents from Pomace.

Abstract

pomace accounts for 20% of the fruit's weight that is not utilized when it is juiced. The pomace contains bioactive phenolic substances such as anthocyanins and flavonoids. To improve the utilization value of pomace, an optimized extraction method for the residual polyphenols was developed using response surface methodology (RSM). The resulting extract was analyzed by high performance liquid chromatography (HPLC), and the in vitro hypoglycemic activity and antioxidant activity of the polyphenolic compounds obtained were also investigated. The optimum extraction conditions (yielding 24.37 mg·g total polyphenols content) were: extraction temperature 60 °C, ultrasonic power 270 W, ethanol concentration 53%, extraction time 57 min, and solid to liquid ratio 1:34. Four polyphenolic compounds were identified in the pomace extract by HPLC: myricitrin, cyanidin--glucoside, hyperoside, and quercitrin. DPPH and hydroxyl radical scavenging tests showed that the polyphenols extract had strong antioxidant abilities. It is evident that the residual polyphenols present in pomace have strong hypoglycemic activity and the juiced fruits can be further exploited for medicinal purposes.

废渣占水果榨汁后未被利用部分的 20%。废渣中含有生物活性酚类物质,如花青素和类黄酮。为了提高废渣的利用价值,采用响应面法(RSM)开发了一种优化的剩余多酚提取方法。采用高效液相色谱法(HPLC)对所得提取物进行分析,并研究了所得多酚化合物的体外降血糖活性和抗氧化活性。最佳提取条件(总多酚含量 24.37mg·g)为:提取温度 60°C,超声功率 270W,乙醇浓度 53%,提取时间 57min,固液比 1:34。HPLC 鉴定废渣提取物中含有 4 种多酚类化合物:杨梅素、矢车菊素-葡萄糖苷、金丝桃苷和槲皮素。DPPH 和羟基自由基清除试验表明,多酚提取物具有较强的抗氧化能力。显然,废渣中存在的残留多酚具有很强的降血糖活性,榨汁后的水果可进一步开发药用。
