Abstract

OBJECTIVE

To explore the effect of the Wnt/-catenin signaling pathway on the proliferation and apoptosis of gastric cancer cells.

METHODS

An MTT colorimetric assay was used to detect the inhibitory effect of the Wnt/-catenin signaling pathway inhibitor FH535 on the proliferation of MKN45 gastric cancer cells. The cell proliferation index (PI) and apoptosis index (AI) were measured by flow cytometry. The expression levels of -catenin, c-myc, and cleaved caspase-3 in MKN45 gastric cancer cells were detected.

RESULTS

After using the Wnt/-catenin signaling pathway inhibitor FH535, MKN45 gastric cancer cells showed obvious shrinkage, death, and cell density decrease. MTT showed that the A value of MKN45 gastric cancer cells in FH535 group was significantly lower than that in the control group ( < 0.05). The survival rate of MKN45 gastric cancer cells in FH535 group was significantly lower than that in the control group ( < 0.05). The cell cycle of gastric cancer was arrested in 0/1 phase. The expression levels of -catenin and c-myc protein in MKN45 gastric cancer cells in FH535 group decreased significantly ( < 0.05), while the expression level of cleaved caspase-3 protein increased significantly ( < 0.05).

CONCLUSION

The Wnt/-catenin signal molecule can maintain the proliferation of gastric cancer cells. Inhibition of the Wnt/-catenin signaling pathway can inhibit the proliferation of gastric cancer cells and promote the apoptosis of MKN45 gastric cancer cells.

目的

探讨 Wnt/-catenin 信号通路对胃癌细胞增殖和凋亡的影响。

方法

采用 MTT 比色法检测 Wnt/-catenin 信号通路抑制剂 FH535 对 MKN45 胃癌细胞增殖的抑制作用。流式细胞术检测细胞增殖指数(PI)和凋亡指数(AI)。检测 MKN45 胃癌细胞中-catenin、c-myc 和 cleaved caspase-3 的表达水平。

结果

使用 Wnt/-catenin 信号通路抑制剂 FH535 后,MKN45 胃癌细胞明显皱缩、死亡、细胞密度降低。MTT 结果显示,FH535 组 MKN45 胃癌细胞的 A 值明显低于对照组( < 0.05)。FH535 组 MKN45 胃癌细胞的存活率明显低于对照组( < 0.05)。胃癌细胞周期停滞在 0/1 期。FH535 组 MKN45 胃癌细胞中-catenin 和 c-myc 蛋白表达水平明显降低( < 0.05),而 cleaved caspase-3 蛋白表达水平明显升高( < 0.05)。

结论

Wnt/-catenin 信号分子可维持胃癌细胞的增殖。抑制 Wnt/-catenin 信号通路可抑制胃癌细胞的增殖,促进 MKN45 胃癌细胞的凋亡。
