Abstract

The new polyketides lopouzanones A and B, as well as the new 1--acetyl and 2--acetyl derivatives of dendrodochol B, were isolated from the sponge-derived marine fungus strain 168CLC-57.3. Moreover, six known polyketides, gliorosein, balticolid, dendrodolide G, dihydroisocoumarine, (-)-5-methylmellein, and dendrodochol B, were identified. The structures of the isolated compounds were determined by a combination of NMR and ESIMS techniques. The absolute configurations of the lopouzanones A and B were determined using the Mosher's method. The cytotoxicity of the isolated compounds against human prostate cancer cells PC-3 and normal rat cardiomyocytes H9c2 was investigated. Gliorosein showed weak DPPH radical-scavenging activity and in vitro cardioprotective effects toward rotenone toxicity and CoCl-mimic hypoxia.

新的聚酮类化合物 lopouzanones A 和 B,以及 dendrodochol B 的 1--乙酰基和 2--乙酰基衍生物,是从海绵来源的海洋真菌 168CLC-57 中分离得到的。此外,还鉴定了六种已知的聚酮类化合物,gliorosein、balticolid、dendrodolide G、二氢异香豆素、(-)-5-甲基麦角甾醇和 dendrodochol B。通过 NMR 和 ESIMS 技术的组合确定了分离化合物的结构。lopouzanones A 和 B 的绝对构型是通过 Mosher 法确定的。对分离得到的化合物对人前列腺癌细胞 PC-3 和正常大鼠心肌细胞 H9c2 的细胞毒性进行了研究。Gliorosein 表现出较弱的 DPPH 自由基清除活性和对鱼藤酮毒性和 CoCl 模拟缺氧的体外心脏保护作用。
