The success of dental implants is determined by the osteointegration process. Many studies state that smoking cigarettes can inhibit osseointegration, but the inhibition mechanism is still unclear.The aim of this study was to identify and analyze the effect of nicotine on the inhibition of dental implant osseointegration through the expression of nicotinic acetylcholine receptor (nAChR), nuclear factor of activated T cells cytoplasmic 1 (NFATc1), osteoclast, and osteoblast numbers.
This study is an experimental study of 16 New Zealand rabbits, randomized across two groups. Group 1 (eight rabbits) was a control group, and group 2 (eight rabbits) was a treatment group. The treatment group was given 2.5 mg/kg body weight/day of nicotine by injection 1 week before placement of the implant until the end of research. Observations were made in the first and the eighth week by measuring the number of osteoblast and osteoclast by immunohistology test and the expression of nAChR and NFATc1 by immunohistochemistry test.
Data was analyzed using a one-way analysis of variance and Student's -test. A -value of < 0.05 was considered statistically significant.
Significant differences were found between the control and treatment groups ( < 0.05). Results showed that nicotine increases the expression of nAChR and decreases the number of osteoblasts and the expression of BMP2 and osteocalcin.
Nicotine inhibits the osseointegration of dental implants by increasing nAChR, NFATc1, osteoclast numbers, and decreasing osteoblast numbers.
牙种植体的成功取决于骨整合过程。许多研究表明吸烟会抑制骨整合,但其抑制机制仍不清楚。本研究的目的是通过烟碱型乙酰胆碱受体(nAChR)、活化T细胞核因子细胞质1(NFATc1)、破骨细胞和成骨细胞数量的表达来识别和分析尼古丁对牙种植体骨整合抑制的影响。
本研究是一项对16只新西兰兔的实验研究,随机分为两组。第1组(8只兔)为对照组,第2组(8只兔)为治疗组。治疗组在植入种植体前1周开始每天注射2.5mg/kg体重的尼古丁,直至研究结束。在第1周和第8周通过免疫组织学检测测量成骨细胞和破骨细胞数量,并通过免疫组织化学检测观察nAChR和NFATc1的表达。
数据采用单因素方差分析和Student's -检验进行分析。P值<0.05被认为具有统计学意义。
对照组和治疗组之间存在显著差异(P<0.05)。结果表明,尼古丁增加了nAChR的表达,减少了成骨细胞数量以及骨形态发生蛋白2(BMP2)和骨钙素的表达。
尼古丁通过增加nAChR、NFATc1、破骨细胞数量和减少成骨细胞数量来抑制牙种植体的骨整合。
