Abstract

OBJECTIVE

To investigate the effect of salidroside on the exercise tolerance of mice under high altitude hypoxia environment.

METHODS

C57BL/6J healthy male mice were randomly divided into normoxia control group, model control group, capsule group and salidroside low-dose (5 mg/kg), medium-dose (10 mg/kg) and high-dose (20 mg/kg) groups, with 15 mice in each group. After 3 days, all groups (except the normoxia control group) entered a plateau with an altitude of 4010 m. After 1 day of hypoxia exposure, the exhausted swimming test was performed to determine the exhaustive time of mice; the pathological changes of liver and muscle tissue were observed with hematoxylin and eosin staining. The levels of malondialdehyde (MDA), hydrogen peroxide (H O ), glutathione (GSH), total superoxide dismutase (T-SOD), glycogen, lactate and ATPase were measured and compared among groups.

RESULTS

Compared with the normoxia control group, the exhaustive swimming time of the model control group was shortened ( <0.05), the liver tissue and muscle tissue were pathologically damaged, the level of oxidative stress was significantly increased, the levels of sodium potassium ATPase and calcium magnesium ATPase were significantly increased. Compared with the model control group, the exhaustive swimming time of the mice in the capsule group and salidroside groups was significantly prolonged ( <0.05). The oxidative stress injury was alleviated, the contents of MDA, H O and lactic acid in liver and muscle tissues decreased, the contents of GSH, liver glycogen and muscle glycogen increased, and the activities of T-SOD and ATPase increased (all <0.05).

CONCLUSION

Salidroside has significant anti-fatigue activity, and its anti-fatigue effect is related to the reduction of oxidative stress damage, the reduction of the accumulation of undesirable metabolites and the increase in the reserve of energy substances.

目的

研究红景天苷对高原缺氧环境下小鼠运动耐力的影响。

方法

将 C57BL/6J 雄性健康小鼠随机分为常氧对照组、模型对照组、胶囊组及红景天苷低(5mg/kg)、中(10mg/kg)、高(20mg/kg)剂量组,每组 15 只。除常氧对照组外,各组于第 3 天进入海拔 4010m 的高原,暴露 1 天后进行力竭游泳实验,测定小鼠的力竭游泳时间;采用苏木精-伊红(HE)染色观察肝、肌肉组织的病理变化。测定各组丙二醛(MDA)、过氧化氢(H O )、还原型谷胱甘肽(GSH)、总超氧化物歧化酶(T-SOD)、糖原、乳酸及三磷酸腺苷酶(ATPase)的水平并进行比较。

结果

与常氧对照组比较,模型对照组小鼠的力竭游泳时间缩短( <0.05),肝、肌肉组织病理损伤,氧化应激水平显著升高,钠钾三磷酸腺苷酶和钙镁三磷酸腺苷酶水平显著升高;与模型对照组比较,胶囊组及红景天苷各剂量组小鼠的力竭游泳时间显著延长( <0.05),氧化应激损伤减轻,肝、肌肉组织中 MDA、H O 及乳酸含量降低,GSH、肝糖原及肌糖原含量升高,T-SOD、ATPase 活性升高(均 <0.05)。

结论

红景天苷具有显著的抗疲劳作用,其抗疲劳作用与减轻氧化应激损伤、减少有害物质蓄积、增加能量物质储备有关。
