Somatic cell hybrids between Sp2/O-Ag14 mouse myeloma cells and lymphocytes derived from BALB/c mice hyperimmunized with sheep red blood cells (SRBC) were produced. One hybrid producing IgG1 antibody to SRBC was selected, cloned twice and subsequently transferred to BALB/c mice. After a number of transfers it was found that the antibody titer in ascites fluid gradually decreased. Cytogenetic analysis revealed gradual chromosome loss in the hybrid clone, which produced progressively less antibody.
构建了Sp2/O-Ag14小鼠骨髓瘤细胞与用绵羊红细胞(SRBC)超免疫的BALB/c小鼠来源的淋巴细胞之间的体细胞杂种。选择了一个产生抗SRBC的IgG1抗体的杂种,进行了两次克隆,随后转移到BALB/c小鼠体内。经过多次传代后发现,腹水液中的抗体效价逐渐降低。细胞遗传学分析显示杂种克隆中的染色体逐渐丢失,产生的抗体也越来越少。
