Abstract

A Gram-stain-negative, aerobic, non-spore-forming, non-motile, coccus-shaped, and red-pigmented bacterial strain designated as CJ14 was isolated from lettuce cultivation soil in Yong-In, South Korea. Strain CJ14 grew optimally on Luria-Bertani agar at 37 ℃ and pH 7.0 in the absence of NaCl. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain CJ14 belonged to the genus Rhodoligotrophos in the family Rhodoligotrophaceae and was closely related to Rhodoligotrophos defluvii lm1 (98.4% similarity). The genome size of strain CJ14 was 4.8 Mbp consisting of a single contig, and the DNA G + C content of strain CJ14 was 61.8%. The genomic comparison of strain CJ14 to R. defluvii lm1 showed an average nucleotide identity value of 77.9%, and the digital DNA-DNA hybridization value was 22.4%. Genomic analyses revealed that strain CJ14 possessed a gene cluster for ochrobactin biosynthesis, highly homologous to the siderophore gene cluster from Nitratireductor thuwali Nit1536. Strain CJ14 contained ubiquinone (Q-10) as the predominant respiratory quinone. The major cellular fatty acids of strain CJ14 were C C cyclo ω8c and summed feature 8. The polar lipid profile was composed of phosphatidylethanolamine, phosphatidylglycerol, phosphatidylcholine, diphosphatidylglycerol, one unidentified aminolipid, one unidentified glycolipid and five unidentified lipids. Based on polyphasic taxonomy study, strain CJ14 could be classified as a novel species of the genus Rhodoligotrophos, for which the name Rhodoligotrophos ferricapiens sp. nov. is proposed. The type strain is CJ14 (= KACC 23063 = JCM 36057).

从韩国龙仁市生菜种植土壤中分离出一株革兰氏染色阴性、需氧、不形成芽孢、无运动性、球形且呈红色色素的细菌菌株,命名为CJ14。菌株CJ14在不含氯化钠的情况下,于37℃、pH 7.0的Luria-Bertani琼脂培养基上生长最佳。基于16S rRNA基因序列的系统发育分析表明,菌株CJ14属于红寡养菌科的红寡养菌属,与脱氮红寡养菌lm1密切相关(相似度为98.4%)。菌株CJ14的基因组大小为4.8 Mbp,由一个重叠群组成,其DNA G+C含量为61.8%。菌株CJ14与脱氮红寡养菌lm1的基因组比较显示,平均核苷酸同一性值为77.9%,数字DNA-DNA杂交值为22.4%。基因组分析表明,菌株CJ14拥有一个与来自硫还原硝酸盐杆菌Nit1536的铁载体基因簇高度同源的奥罗巴菌素生物合成基因簇。菌株CJ14含有泛醌(Q-10)作为主要呼吸醌。菌株CJ14的主要细胞脂肪酸为C C环ω8c和总和特征8。极性脂质谱由磷脂酰乙醇胺、磷脂酰甘油、磷脂酰胆碱、二磷脂酰甘油、一种未鉴定的氨基脂质、一种未鉴定的糖脂和五种未鉴定的脂质组成。基于多相分类学研究,菌株CJ14可被归类为红寡养菌属的一个新物种,为此提出了新名称铁嗜红寡养菌(Rhodoligotrophos ferricapiens sp. nov.)。模式菌株为CJ14(=KACC 23063=JCM 36057)。
