Biostability is a key feature for DNA nanostructures to be used in biological applications. With several strategies being employed to enhance the nuclease resistance of DNA nanostructures, methods to analyze the nuclease degradation are also needed. This chapter describes a protocol to evaluate the nuclease resistance of DNA motifs and nanostructures using polyacrylamide gel electrophoresis.
