Abstract

A Gram-stain-negative, rod-shaped and non-motile bacterium, designated as strain SYSU DXS3258, was isolated from red sandy conglomerate sample collected from the Danxia Mountain in Guangdong Province, PR China. The bacterium was found to grow optimally at 28 °C, pH 7.0, and with 0-1% (w/v) NaCl. Phylogenetic analyses based on the 16S rRNA gene sequences placed strain SYSU DXS3258 within the family Sphingobacteriaceae. Strain SYSU DXS3258 showed the highest 16S rRNA gene sequence similarity to Pararcticibacter tournemirensis TF5-37.2-LB10 (95.5%), Paradesertivirga mongoliensis 1-32 (95.3%) and Desertivirga xinjiangensis 12157 (95.2%). Digital DNA-DNA hybridization, orthologous average nucleotide identity and average amino acid identity values between strain SYSU DXS3258 and the closest related members within the Sphingobacteriaceae were ≤ 13%, 70.3% and 69.9%, respectively. The genomic DNA G + C content was 38.8%. The major fatty acid was iso-C, C and C 2-OH. The predominant polar lipids were phosphatidylethanolamine, three glycolipids, one phospholipid and one unidentified polar lipid. The major respiratory quinone was Q-7. Based on phenotypic, phylogenetic, chemotaxonomic and genomic results, we propose that strain SYSU DXS3258 represents a novel species of a novel genus within the family Sphingobacteriaceae, for which the name Rubrolithibacter danxiaensis gen. nov., sp. nov. was proposed. The type strain is SYSU DXS3258 (= MCCC 1K09420 = KCTC 102322).

从中国广东省丹霞山采集的红色砂质砾岩样本中分离出一株革兰氏阴性、杆状且无运动性的细菌,命名为菌株SYSU DXS3258。该细菌在28°C、pH 7.0以及0-1%(w/v)NaCl条件下生长最佳。基于16S rRNA基因序列的系统发育分析将菌株SYSU DXS3258置于鞘脂杆菌科内。菌株SYSU DXS3258与图尔内米副北极杆菌TF5-37.2-LB10(95.5%)、蒙古拟沙漠杆菌1-32(95.3%)和新疆沙漠杆菌12157(95.2%)的16S rRNA基因序列相似性最高。菌株SYSU DXS3258与鞘脂杆菌科内最密切相关成员之间的数字DNA-DNA杂交、直系同源平均核苷酸同一性和平均氨基酸同一性值分别≤13%、70.3%和69.9%。基因组DNA的G+C含量为38.8%。主要脂肪酸为异-C、C和C 2-OH。主要极性脂为磷脂酰乙醇胺、三种糖脂、一种磷脂和一种未鉴定的极性脂。主要呼吸醌为Q-7。基于表型、系统发育、化学分类和基因组结果,我们提议菌株SYSU DXS3258代表鞘脂杆菌科内一个新属的一个新物种,为此提出新属名为丹霞红岩杆菌属(Rubrolithibacter),新种名为丹霞丹霞红岩杆菌(Rubrolithibacter danxiaensis)。模式菌株为SYSU DXS3258(=MCCC 1K09420=KCTC 102322)。
