A novel actinomycete designated CME 23 was isolated from cassava rhizosphere soil collected from Chon Buri province, Thailand. The strain was characterized using a polyphasic approach and classified as a member of the genus Streptomyces. Strain CME 23 formed spiral chains of non-motile cylindrical spores with a smooth surface developed on aerial mycelia. The cell wall peptidoglycan contained LL-diaminopimelic acid. Rhamnose, ribose, mannose, and glucose were found in its whole-cell hydrolysates. The predominant menaquinones were MK-9(H) and MK-9(H). The predominant fatty acids were iso-C, anteiso-C, and iso-C. The polar lipid profile consisted of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannoside, and three unidentified phospholipids. The 16S rRNA gene sequence analysis indicated that strain CME 23 shared the highest sequence similarity with Streptomyces curacoi DSM 40107 (98.8%). The average amino acid identity (ANI) values and digital DNA-DNA hybridization (dDDH) values between the genome sequence of strain CME 23 and closest type strains of Streptomyces curacoi DSM 40107, Streptomyces cylindrosporus 7R015, and Streptomyces shaanxiensis CCNWHQ 0031, were 83.0-88.5% and 28.0-32.8%, respectively. The G + C content of the genome DNA was 71.1%. Based on comparative analyses of genotypic and phenotypic data, the actinomycete strain CME 23 (= TBRC 15810 = NBRC 115859) represents a novel Streptomyces species, for which the name Streptomyces gilvus sp. nov. is proposed. The in vitro assay showed that strain CME 23 produced IAA, siderophore, as well as solubilize phosphate and zinc.
从泰国春武里府采集的木薯根际土壤中分离出一株新的放线菌,命名为CME 23。采用多相分类法对该菌株进行了表征,并将其归类为链霉菌属的一员。菌株CME 23在气生菌丝体上形成表面光滑的非运动性圆柱形孢子的螺旋链。细胞壁肽聚糖含有LL-二氨基庚二酸。在其全细胞水解产物中发现了鼠李糖、核糖、甘露糖和葡萄糖。主要的甲基萘醌是MK-9(H)和MK-9(H)。主要脂肪酸为异-C、anteiso-C和异-C。极性脂质谱由双磷脂酰甘油、磷脂酰乙醇胺、磷脂酰甘油、磷脂酰肌醇、磷脂酰肌醇甘露糖苷和三种未鉴定的磷脂组成。16S rRNA基因序列分析表明,菌株CME 23与库拉索链霉菌DSM 40107的序列相似性最高(98.8%)。菌株CME 23的基因组序列与库拉索链霉菌DSM 40107、圆柱形链霉菌7R015和陕西链霉菌CCNWHQ 0031的最接近模式菌株之间的平均氨基酸同一性(ANI)值和数字DNA-DNA杂交(dDDH)值分别为83.0-88.5%和28.0-32.8%。基因组DNA的G + C含量为71.1%。基于基因型和表型数据的比较分析,放线菌菌株CME 23(= TBRC 15810 = NBRC 115859)代表一个新的链霉菌物种,为此提出了吉尔维链霉菌新种的名称。体外试验表明,菌株CME 23产生吲哚乙酸、铁载体,还能溶解磷酸盐和锌。
