Abstract

We observed the rules of drug release in vitro from a liver targeting preparation-mitoxantrone polybutylcyanoacrylate nanospheres freeze-drying injection by dynamic penetration system and spectrophotometry. The results showed that the drug release of this nanospheres preparation accorded with the two phases kinetics rules in vitro. In the process of drug release, there were two phases. The first phase was a quick release phase, the quantity of released drug within 4 hours being more than 40%, t1/2 alpha = 1.40h. The second phase was a slow release phase, t1/2 beta = 169.02h. The rules of drug release accord with the normal rules of drug use. The preparation may be clinically used.

我们通过动态渗透系统和分光光度法观察了一种肝靶向制剂——米托蒽醌聚氰基丙烯酸正丁酯纳米球冻干注射剂的体外释药规律。结果表明,该纳米球制剂的药物释放体外符合双相动力学规律。在药物释放过程中,存在两个阶段。第一阶段为快速释放阶段,4小时内释放药量超过40%,t1/2α = 1.40小时。第二阶段为缓慢释放阶段,t1/2β = 169.02小时。药物释放规律符合药物使用的常规规律。该制剂可临床应用。
