The role of a chymotrypsin-like enzyme in the release of histamine in anaphylaxis has been investigated using sensitized rat peritoneal mast cells. Inhibition of this enzyme did not inhibit histamine release and, conversely, when histamine release was blocked the esterase activity was unaffected. It is concluded that the active chymotrypsin-like enzyme in these cells is not involved in the anaphylactic reaction. One of the inhibitors used (DFP) produced a dual action on histamine release. A possible explanation is discussed.
