To develop a reversed-phase HPLC method for simultaneous determination of gastrodin, adenosin, 4-hydroxybenzyl alcohol and 4-hydroxybenzaldehyde in Rhizoma Gastrodia.
A Kromasil C18 column (4.6 mm x 250 mm, 5 microm) was used with a methanol-water-0.1% acetic acid gradient elution system. The eluates were detected at 270 nm, the flow rate was 1.0 mL x min(-1) and the column temperature was 35 degrees C.
The linear range of gastrodin, adenosin, 4-hydroxybenzyl alcohol and 4-hydroxybenzaldehyde were 19.1-383 (r = 0.999 9), 0.620-12.4 (r = 0.999 9), 2.45-49.0 (r = 0.999 9), 0.280-5.63 mg x L(-1) (r = 0.999 6), respectively. The average recoveries (n = 9) of the four components were 96.7% -97.7%, RSD < 1.6%.
The method is accurate, sensitive and reliable for determination of gastrodin, adenosin, 4-hydroxybenzyl alcohol and 4-hydroxybenzaldehyde in Rhizoma Gastrodia.
建立一种反相高效液相色谱法同时测定天麻中天麻素、腺苷、4-羟基苄醇和4-羟基苯甲醛的含量。
采用Kromasil C18柱(4.6 mm×250 mm,5μm),以甲醇-水-0.1%乙酸梯度洗脱体系进行洗脱。在270 nm波长处检测洗脱液,流速为1.0 mL·min⁻¹,柱温为35℃。
天麻素、腺苷、4-羟基苄醇和4-羟基苯甲醛的线性范围分别为19.1 - 383(r = 0.999 9)、0.620 - 12.4(r = 0.999 9)、2.45 - 49.0(r = 0.999 9)、0.280 - 5.63 mg·L⁻¹(r = 0.999 6)。4种成分的平均回收率(n = 9)为96.7% - 97.7%,相对标准偏差(RSD)< 1.6%。
该方法用于测定天麻中天麻素、腺苷、4-羟基苄醇和4-羟基苯甲醛准确、灵敏、可靠。
