To investigate the correlation between E670G polymorphism of proprotein convertase subtilisin/kexin type 9 (PCSK9) gene and coronary heart disease (CHD), and contrastively study the regional differences of E670G polymorphism of PCSK9 gene between patients with CHD among the Han population in Hainan and three provinces in the northeast of China (TPNC), providing scientific basis for prevention and treatment of patients with CHD in different regions.
A total of 233 cases of patients with CHD were selected from the Han population in Hainan and TPNC as the experimental group (118 cases from Hainan, 115 cases from TPNC), and 239 cases with non-CHD were selected among the Han population also in the two regions as control group (125 cases from Hainan, 114 cases from TPNC). The triglyceride (TG), total cholesterol (TC), high density lipoprotein cholesterol and low density lipoprotein cholesterol (LDL-C) levels of plasma were tested and PCR-RFLP method was used to test the E670G polymorphism of PCSK9 gene. The statistical software package SPSS 21.0 was used for the statistical analysis and P < 0.05 was considered as statistically significant.
The levels of systolic pressure, diastolic blood pressure, fasting blood sugar, TC, TG, and LDL-C of patients in CHD group were significantly higher than those in non-CHD group, while the high density lipoprotein cholesterol level was lower than that in non-CHD group (P < 0.05). In CHD group, the frequencies of AG, GG genotypes of PCSK9 gene and G allele were higher than those in non-CHD group (P < 0.05), and in CHD group, the frequencies of AG, GG genotypes and G allele of patients both in Hainan and TPNC were higher than those in control group (P < 0.05). Among the patients with CHD, the frequencies of GG genotype and G allele of patients in Hainan were lower than those in TPNC (P < 0.05), and in CHD group, the levels of TG, TC and LDL-C of GG genotype were higher than those of AA genotype (P < 0.05). While in non-CHD group, there were no significant differences between the frequencies of GG genotype and G allele of patients in Hainan and TPNC (P > 0.05).
There was a close correlation between the E670G polymorphism of PCSK9 gene and CHD with serum lipid level. Among Han population in Hainan and TPNC, the E670G polymorphism of PCSK9 gene of patients with CHD exhibited regional differences.
探讨前蛋白转化酶枯草溶菌素 9(PCSK9)基因 E670G 多态性与冠心病(CHD)的相关性,并对比研究中国东北三省(TPNC)与海南汉族人群中 CHD 患者 PCSK9 基因 E670G 多态性的地域差异,为不同地区 CHD 患者的防治提供科学依据。
选取海南及 TPNC 汉族人群中的 233 例 CHD 患者作为实验组(海南 118 例,TPNC 115 例),同时选取海南及 TPNC 汉族人群中的 239 例非 CHD 患者作为对照组(海南 125 例,TPNC 114 例)。检测血浆甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇和低密度脂蛋白胆固醇(LDL-C)水平,并采用 PCR-RFLP 法检测 PCSK9 基因 E670G 多态性。采用 SPSS 21.0 统计软件包进行统计学分析,P<0.05 为差异有统计学意义。
CHD 组患者的收缩压、舒张压、空腹血糖、TC、TG、LDL-C 水平明显高于非 CHD 组,而高密度脂蛋白胆固醇水平明显低于非 CHD 组(P<0.05)。在 CHD 组中,PCSK9 基因 AG、GG 基因型和 G 等位基因的频率明显高于非 CHD 组(P<0.05),且在海南和 TPNC 的 CHD 组患者中,AG、GG 基因型和 G 等位基因的频率明显高于对照组(P<0.05)。在 CHD 患者中,海南患者的 GG 基因型和 G 等位基因频率明显低于 TPNC 患者(P<0.05),且在 CHD 组中,GG 基因型患者的 TG、TC 和 LDL-C 水平明显高于 AA 基因型患者(P<0.05)。而非 CHD 组中,海南和 TPNC 患者的 GG 基因型和 G 等位基因频率无明显差异(P>0.05)。
PCSK9 基因 E670G 多态性与 CHD 及其血脂水平密切相关。在海南和 TPNC 的汉族人群中,CHD 患者的 PCSK9 基因 E670G 多态性存在地域差异。
