A Gram-positive, aerobic, non-motile, rod-shapeds, catalase-positive, and oxidase-negative strain, designated Y49, was isolated from sewage collected from Jilin Agricultural University, China. It grew at 20-40°C (optimum at 30°C), at pH 6.0-8.0 (optimum at 7.0) and at 0-1.0% sodium chloride (optimum at 0%). The major isoprenoid quinone was menaquinone-8 (MK-8) and the polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylmethylethanolamine, four unidentified lipids, and two unidentified aminolipids. The peptidoglycan was meso-diaminopimelic acid. The cell-wall sugars were galactose, arabinose, and glucose. The fatty acids were C, C, C ω9c, C ω9c, C ω6c (6,9,12), C ω9c, and C. The DNA G+C content was 51.4 mol%. Based on the 16S rRNA gene sequence analysis, the nearest phylogenetic neighbors of strain Y49 were Corynebacterium efficiens DSM 44549 (97.5%), Corynebacterium callunae DSM 20147 (97.2%), Corynebacterium deserti GIMN 1.010 (96.8%), Corynebacterium glutamicum ATCC 13032 (96.4%), and other species belonging to this genus (92.3-95.4%). The DNA-DNA relatedness value between strain Y49 and C. efficiens DSM 44549, C. callunae DSM 20147, C. deserti GIMN1.010, and C. glutamicum ATCC 13032 was 25.5±2.0%, 21.1±1.0%, 16.5±0.5%, and 13.5±0.9%, respectively. Based on the phylogenetic analysis, chemotaxonomic data, physiological characteristics and DNA-DNA hybridization data, strain Y49 represents a novel species of the genus Corynebacterium, for which the name Corynebacterium defluvii sp nov. is proposed. The type strain is Y49 (= KCTC 39731 =CGMCC 1.15506).
从中国吉林农业大学采集的污水中分离出一株革兰氏阳性、需氧、不运动、杆状、过氧化氢酶阳性且氧化酶阴性的菌株,命名为Y49。它在20-40°C(最适温度30°C)、pH 6.0-8.0(最适pH 7.0)和0-1.0%氯化钠(最适浓度0%)的条件下生长。主要类异戊二烯醌为甲基萘醌-8(MK-8),极性脂为双磷脂酰甘油、磷脂酰甘油、磷脂酰甲基乙醇胺、四种未鉴定的脂类和两种未鉴定的氨基脂类。肽聚糖为内消旋二氨基庚二酸。细胞壁糖类为半乳糖、阿拉伯糖和葡萄糖。脂肪酸为C、C、C ω9c、C ω9c、C ω6c(6,9,12)、C ω9c和C。DNA G+C含量为51.4 mol%。基于16S rRNA基因序列分析,菌株Y49最近的系统发育邻株为高效棒状杆菌DSM 44549(97.5%)、紫红红球菌DSM 20147(97.2%)、沙漠棒状杆菌GIMN 1.010(96.8%)、谷氨酸棒状杆菌ATCC 13032(96.4%)以及该属的其他物种(92.3-95.4%)。菌株Y49与高效棒状杆菌DSM 44549、紫红红球菌DSM 20147、沙漠棒状杆菌GIMN1.010和谷氨酸棒状杆菌ATCC 13032之间的DNA-DNA相关性值分别为25.5±2.0%、21.1±1.0%、16.5±0.5%和13.5±0.9%。基于系统发育分析、化学分类数据、生理特征和DNA-DNA杂交数据,菌株Y49代表棒状杆菌属的一个新物种,为此提出新名称污水棒状杆菌(Corynebacterium defluvii sp nov.)。模式菌株为Y49(=KCTC 39731 =CGMCC 1.15506)。
