Nakai ex F. Maek (VD) has been used as traditional medicine for the treatment of restlessness and sleeping disorders. However, it is still unclear whether obesity in mice can be altered by diet supplementation with VD. In this study, we first investigated the influences of VD on the accumulation of lipid content in 3T3-L1 cells; and the results showed that the above-ground VD extracts (VDAE) suppressed the differentiation of 3T3-L1 preadipocytes in a concentration-dependent manner without cytotoxicity. Thus, the effects of VDAE on preventing obesity were then studied in the C57BL/6N mice for 10 weeks ( = 6): normal-fat diet, high-fat diet (HFD), HFD supplemented with 1% (10 g/kg) extract (positive control), and HFD supplemented with 1% (10 g/kg) VDAE. The results showed that VDAE reduced food efficiency ratio, body weight, epididymal adipose and hepatic tissue weight, hepatic lipid metabolites, and triacylglycerol and cholesterol serum levels compared to the high-fat diet group. Moreover, VD significantly inhibited the expression of adipogenic genes, such as , , and , and lipogenic genes, such as , , , and , in epididymal adipose tissue and hepatic tissue. These findings indicate anti-adipogenic and anti-lipogenic effects of VDAE and suggest that it could be a potent functional food ingredient for the prevention of high-fat diet-induced obesity.
中井提取物(VD)已被用作治疗烦躁不安和睡眠障碍的传统药物。然而,目前尚不清楚通过饮食补充VD是否能改变小鼠的肥胖状况。在本研究中,我们首先研究了VD对3T3-L1细胞脂质含量积累的影响;结果表明,地上部分的VD提取物(VDAE)以浓度依赖的方式抑制3T3-L1前脂肪细胞的分化,且无细胞毒性。因此,随后在C57BL/6N小鼠中研究了VDAE预防肥胖的效果,为期10周(每组n = 6):正常脂肪饮食、高脂饮食(HFD)、补充1%(10 g/kg)提取物的高脂饮食(阳性对照)以及补充1%(10 g/kg)VDAE的高脂饮食。结果显示,与高脂饮食组相比,VDAE降低了食物效率比、体重、附睾脂肪和肝脏组织重量、肝脏脂质代谢产物以及甘油三酯和胆固醇血清水平。此外,VD显著抑制了附睾脂肪组织和肝脏组织中脂肪生成基因(如 、 、 )以及脂质生成基因(如 、 、 、 )的表达。这些发现表明VDAE具有抗脂肪生成和抗脂质生成作用,并提示它可能是预防高脂饮食诱导肥胖的一种有效功能性食品成分。
