Abstract

Strains pyc13 and ZGT13 were isolated from Lake Pengyan and Lake Zigetang on Tibetan Plateau, respectively. Both strains were Gram-negative, catalase- and oxidase-positive, aerobic, rod-shaped, nonmotile, and nonflagellated bacteria. Phylogenetic analysis based on 16S rRNA gene sequences showed that strains pyc13 and ZGT13 belong to the genus Halomonas, with Halomonas alkalicola 56-L4-10aEn as their closest neighbor, showing 97.4% 16S rRNA gene sequence similarity. The predominant respiratory quinone of both strains was Q-9, with Q-8 as a minor component. The major fatty acids of both strains were Cω6c/Cω7c, Cω6c/Cω7c, C, and C 3OH. The polar lipids of both strains consisted of phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol, glycolipid, phospholipids of unknown structure containing glucosamine, and unidentified phospholipids. The DNA G + C content of pyc13 and ZGT13 were 62.6 and 63.4 mol%, respectively. The DNA-DNA hybridization values of strain pyc13 were 34, 41, 61, 35, and 35% with the reference strains H. alkalicola 56-L4-10aEn, H. sediminicola CPS11, H. mongoliensis Z-7009, H. ventosae Al12, and H. fontilapidosi 5CR, respectively. Phenotypic, biochemical, genotypic, and DNA-DNA hybridization data showed that strains pyc13 and ZGT13 represent a new species within the genus Halomonas, for which the name H. tibetensis sp. nov. is proposed. The type strain is pyc13 (= CGMCC 1.15949 = KCTC 52660).

菌株 pyc13 和 ZGT13 分别从青藏高原的澎湖和则哥塘湖分离得到。两株菌均为革兰氏阴性、过氧化氢酶和氧化酶阳性、需氧、杆状、非运动、无鞭毛的细菌。基于 16S rRNA 基因序列的系统发育分析表明,菌株 pyc13 和 ZGT13 属于盐单胞菌属,与盐单胞菌 56-L4-10aEn 最为接近,16S rRNA 基因序列相似度为 97.4%。两株菌的主要呼吸醌均为 Q-9,Q-8 为次要成分。两株菌的主要脂肪酸均为 Cω6c/Cω7c、Cω6c/Cω7c、C 和 C3OH。两株菌的极性脂均由磷脂酰乙醇胺、磷脂酰甘油、双磷脂酰甘油、糖脂、含有葡萄糖胺的未知结构磷脂和未鉴定的磷脂组成。pyc13 和 ZGT13 的 DNA G+C 含量分别为 62.6 和 63.4mol%。菌株 pyc13 与参考菌株 H. alkalicola 56-L4-10aEn、H. sediminicola CPS11、H. mongoliensis Z-7009、H. ventosae Al12 和 H. fontilapidosi 5CR 的 DNA-DNA 杂交值分别为 34、41、61、35 和 35%。表型、生化、基因型和 DNA-DNA 杂交数据表明,菌株 pyc13 和 ZGT13 代表盐单胞菌属的一个新种,命名为西藏盐单胞菌 sp. nov.。模式菌株为 pyc13(=CGMCC 1.15949=KCTC 52660)。
