Abstract

A nitrogen-fixing, endospore-forming bacterium, designated strain L201 was isolated from the leaves of Bryophyllum pinnatum growing in South China Agricultural University. Phylogenetic analysis of the 16S rRNA gene sequence indicated that strain L201 is affiliated with the genus Paenibacillus, and closely related to Paenibacillus albidus Q4-3 (97.4%), Paenibacillus odorifer DSM 15391 (97.3%) and Paenibacillus borealis DSM 13188 (97.2%). The main fatty acids components was anteiso-C (48.1%). The predominant isoprenoid quinone was MK-7. The major polar lipids were found to be diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylglycerol. The G+C content of strain L201 was 43.9%. DNA-DNA relatedness between L201 and the reference strain was 29.8%. Biological and biochemical tests, protein patterns, genomic DNA fingerprinting and comparison of cellular fatty acids distinguished strain L201 from the closely related Paenibacillus species. Based on these data, the novel species Paenibacillus bryophyllum sp. nov. is proposed, with the type strain L201(= KCTC 33951  = GDMCC 1.1251 ).

从华南农业大学生长的落地生根叶片中分离出一株固氮、产芽孢的细菌,命名为L201菌株。16S rRNA基因序列的系统发育分析表明,L201菌株属于类芽孢杆菌属,与白色类芽孢杆菌Q4-3(97.4%)、产香类芽孢杆菌DSM 15391(97.3%)和北方类芽孢杆菌DSM 13188(97.2%)密切相关。主要脂肪酸成分是anteiso-C(48.1%)。主要的类异戊二烯醌是MK-7。发现主要极性脂为双磷脂酰甘油、磷脂酰乙醇胺和磷脂酰甘油。L201菌株的G+C含量为43.9%。L201与参考菌株之间的DNA-DNA相关性为29.8%。生物学和生化试验、蛋白质图谱、基因组DNA指纹图谱以及细胞脂肪酸比较将L201菌株与密切相关的类芽孢杆菌属物种区分开来。基于这些数据,提出了一个新物种——落地生根类芽孢杆菌,模式菌株为L201(=KCTC 33951 =GDMCC 1.1251)。
