Human papilloma virus (HPV) is involved in development of almost all cervical cancers, mainly through the subversion of cellular mechanisms of growth control. Fascin plays central role in subsequent cell transformation events. Fascin mediates stabilization of parallel actin bundles where cellular protrusions are formed; this represents primary stages of cell migration and metastasis. Immunohistochemical assays have shown up-regulation of fascin expression in many epithelial and non-epithelial neoplasms. Therefore, the aim of this study was to investigate HPV infection and fascin expression in samples of cervical cancer. Of 66 patients with confirmed SCC, formalin-fixed specimens, embedded in paraffin blocks were evaluated for HPV infection with nested multiplex polymerase chain reaction (NM-PCR) and for fascin expression with immunohistochemical assays. Statistical analysis was performed using Wilcoxon rank-sum test and SPSS software. A p<0.05 was considered for statistical significance. Of 66 samples, 52 (78.7%) were found positive for HPV infection and fascin over-expression was shown in all squamous cell carcinoma samples. This study showed fascin overexpression in squamous cell carcinoma of the cervix which might be involved in metastasis of cancers induced by some types of HPV, hypothetically through attenuation of inter-cellular adhesions, and induction of cell motility.
人乳头瘤病毒（HPV）几乎与所有宫颈癌的发生都有关，主要是通过破坏细胞生长控制机制。肌动蛋白结合蛋白在随后的细胞转化事件中起核心作用。肌动蛋白结合蛋白介导平行肌动蛋白束的稳定，而细胞突起正是在平行肌动蛋白束处形成；这代表了细胞迁移和转移的初始阶段。免疫组织化学分析表明，在许多上皮性和非上皮性肿瘤中肌动蛋白结合蛋白表达上调。因此，本研究的目的是调查宫颈癌样本中的HPV感染情况和肌动蛋白结合蛋白的表达。在66例确诊为鳞状细胞癌（SCC）的患者中，采用巢式多重聚合酶链反应（NM-PCR）对福尔马林固定、石蜡包埋的标本进行HPV感染评估，并用免疫组织化学分析检测肌动蛋白结合蛋白的表达。使用Wilcoxon秩和检验和SPSS软件进行统计分析。p<0.05被认为具有统计学意义。在66个样本中，52个（78.7%）被发现HPV感染呈阳性，并且在所有鳞状细胞癌样本中均显示肌动蛋白结合蛋白过表达。本研究表明，子宫颈鳞状细胞癌中存在肌动蛋白结合蛋白过表达，这可能参与了某些类型HPV诱导的癌症转移，推测是通过减弱细胞间黏附以及诱导细胞运动来实现的。
