Therapy that can cure osteoarthritis with satisfactory results has not been found to date. In the pathogenesis of osteoarthritis, the genes involved in cartilage degradation include the gene which plays an important role in modulating the occurrence of cartilage damage, which involves activation of pro-inflammatory cytokines. One of the cytokines involved in the cartilage degradation process is Matrix Metalloproteinase (MMP) -13 which is also modulated by NFκβ.
This study aims to look at the expression of the gene and expression of the MMP-13 gene and analyse the relationship of gene expression with MMP-13 gene expression after administration of Mesenchymal Stem Cell Wharton Jelly in synoviocytes in vitro.
This research is pure experimental research. The samples used derived from synovial tissue in osteoarthritis patients who underwent surgery for Total Knee Replacement (TKR). This study was divided into 6 treatment groups with 4 replications. Group I was the synoviocyte OA cell control group which was incubated 24 hours, group II was control of synoviocyte OA cell which was incubated 48 hours, group III was a group of Mesenchymal Stem Cell Wharton Jelly (MSC-WJ) which was incubated 24 hours, group IV was a Mesenchymal Stem Cell Wharton Jelly (MSC-WJ) cell group incubated 48 hours, group V was the co-culture group of synoviocyte-MSC-WJ cells incubated 24 hours and group VI was the co-culture of synoviocyte-MSC-WJ cells which were incubated 48 hours. Observation of gene expression and gene in each group was carried out using qPCR.
The results showed that the analysis of the relationship between gene expression and MMP-13 gene expression in osteoarthritis synoviocytes cells after Mesenchymal Stem Cell Wharton Jelly as big as (r = 0.662).
The conclusion of this study is there was a strong correlation between gene expression and MMP-13 gene expression in osteoarthritis synoviocytes after Mesenchymal Stem Cell Wharton Jelly (r = 0.662).
迄今为止,尚未发现能治愈骨关节炎且效果令人满意的疗法。在骨关节炎的发病机制中,参与软骨降解的基因包括在调节软骨损伤发生中起重要作用的基因,这涉及促炎细胞因子的激活。参与软骨降解过程的细胞因子之一是基质金属蛋白酶(MMP)-13,它也受核因子κB(NFκβ)调节。
本研究旨在观察该基因的表达以及MMP-13基因的表达,并分析体外向滑膜细胞中施用间充质干细胞华通氏胶后该基因表达与MMP-13基因表达之间的关系。
本研究为纯实验性研究。所用样本来自接受全膝关节置换术(TKR)手术的骨关节炎患者的滑膜组织。本研究分为6个治疗组,每组重复4次。第一组是滑膜细胞OA细胞对照组,培养24小时;第二组是滑膜细胞OA细胞对照组,培养48小时;第三组是间充质干细胞华通氏胶(MSC-WJ)组,培养24小时;第四组是间充质干细胞华通氏胶(MSC-WJ)细胞组,培养48小时;第五组是滑膜细胞-MSC-WJ细胞共培养组,培养24小时;第六组是滑膜细胞-MSC-WJ细胞共培养组,培养48小时。使用定量聚合酶链反应(qPCR)对每组中的该基因表达和基因进行观察。
结果显示,在施用间充质干细胞华通氏胶后,骨关节炎滑膜细胞中该基因表达与MMP-13基因表达之间的关系分析结果为(r = 0.662)。
本研究的结论是,在施用间充质干细胞华通氏胶后,骨关节炎滑膜细胞中该基因表达与MMP-13基因表达之间存在强相关性(r = 0.662)。
