Abstract

BACKGROUND

The use of medicinal plants is increasing in several decades for relief many diseases. Indonesia consists of thousands of islands with various plants and the manners of the community using plants as a treatment for every disease traditionally.

AIM

Cytotoxic activity of ethyl acetate fraction (EAF) of fruit was tested towards T47D breast cancer cells.

METHODS

The in vitro cytotoxic activity was performed by MTT assay, and the result was expressed as the IC50 (Inhibitory Concentration), and cell cycle inhibition was investigated by flow cytometry to assess the inhibition in every phase of cell cycle, and the role of expression cyclin D1 and p53 in cell cycle inhibition were performed by immunocytochemistry.

RESULTS

EAF was showed to have high activity with a value of IC 48.94 ± 0.32 µg/mL. EAF of 25 µg/mL caused cell accumulation at G0/G1 (60.48%) and in a control cell (51.69%) and decreased expression of cyclin D1 and increased expression of p53.

CONCLUSION

The results obtained in this study provided scientific support for further investigation on compounds in fruit which in the future could be used for medication.

背景

几十年来,药用植物的使用在不断增加,用于缓解多种疾病。印度尼西亚由数千个岛屿组成,拥有各种各样的植物,并且当地社区传统上有使用植物治疗各种疾病的方式。

目的

测试果实乙酸乙酯部位(EAF)对T47D乳腺癌细胞的细胞毒性活性。

方法

通过MTT法进行体外细胞毒性活性检测,结果以IC50(抑制浓度)表示,通过流式细胞术研究细胞周期抑制情况以评估细胞周期各阶段的抑制作用,并通过免疫细胞化学检测细胞周期蛋白D1和p53表达在细胞周期抑制中的作用。

结果

EAF显示出高活性,IC值为48.94±0.32μg/mL。25μg/mL的EAF导致细胞在G0/G1期积累(60.48%),而对照细胞为(51.69%),同时细胞周期蛋白D1表达降低,p53表达增加。

结论

本研究获得的结果为进一步研究果实中的化合物提供了科学支持,这些化合物未来可能用于药物治疗。
