Angiotensin II (AngII), a major product of renin-angiotensin system (RAS) has important role in induction of hypertension and antihypertensive effect of several medicinal plant was mediated by effect on this agent. Therefore, this study examined the possible effect of hydroalcoholic extract of on hypertension induced by AngII.
Six groups (n = 6) of rats were used as follow: 1) Control, 2) AngII (300 ng/kg), 3) Losartan (Los, 10 mg/kg) + AngII and 4-6) extract (10, 20 & 40 mg/kg,) + AngII. The femoral artery and vein were cannulated for recording cardiovascular parameters and drugs administration, respectively. All drugs were injected intravenously (i.v). Los and all doses of injected 10 min before AngII. Systolic blood pressure (SBP), mean arterial blood pressure (MAP) and heart rate (HR) were recorded throughout the experiment and those peak changes (Δ) were calculated and compared to control and AngII.
AngII significantly increased ΔMAP, ΔSBP and ΔHR than control (P < 0. 01 to P < 0.001) and these increments were significantly attenuated by Los. All doses of significantly reduced peak ΔMAP, ΔSBP, and ΔHR than AngII group (P < 0. 05 to P < 0.001). In addition, peak ΔMAP, ΔSBP in doses 10 and 20 were significant than Los + AngII group (P < 0.05 to P< 0.01) but in dose 40 only MAP was significant (P < 0.05). Peak ΔHR in all doses of C sativus was not significant than Los+ AngII.
Regarding the improving effect of the extract on AngII induced hypertension, it seems that this ameliorating effect partly mediated through inhibition of RAS.
血管紧张素II(AngII)是肾素-血管紧张素系统(RAS)的主要产物,在高血压的诱导中起重要作用,几种药用植物的降压作用是通过对该物质的作用介导的。因此,本研究考察了[植物名称]水醇提取物对AngII诱导的高血压的可能作用。
将六组(n = 6)大鼠按以下方式使用:1)对照组,2)AngII(300 ng/kg)组,3)氯沙坦(Los,10 mg/kg)+ AngII组,以及4 - 6)[植物名称]提取物(10、20和40 mg/kg)+ AngII组。分别插入股动脉和静脉用于记录心血管参数和给药。所有药物均静脉注射(i.v)。Los和所有剂量的[植物名称]提取物在AngII注射前10分钟注射。在整个实验过程中记录收缩压(SBP)、平均动脉压(MAP)和心率(HR),并计算这些峰值变化(Δ),与对照组和AngII组进行比较。
与对照组相比,AngII显著增加了ΔMAP、ΔSBP和ΔHR(P < 0.01至P < 0.001),而Los显著减弱了这些增加。与AngII组相比,所有剂量的[植物名称]提取物均显著降低了峰值ΔMAP、ΔSBP和ΔHR(P < 0.05至P < 0.001)。此外,10和20 mg/kg剂量的提取物的峰值ΔMAP、ΔSBP显著高于Los + AngII组(P < 0.05至P < 0.01),但40 mg/kg剂量时仅MAP有显著差异(P < 0.05)。所有剂量的[植物名称]提取物的峰值ΔHR与Los + AngII组相比无显著差异。
鉴于[植物名称]提取物对AngII诱导的高血压有改善作用,似乎这种改善作用部分是通过抑制RAS介导的。
