Abstract

To investigate the expression level of the maternally expressed gene-3 () of the free long non-coding RNA (lncRNAs) in the plasma of Parkinson's disease (PD) patients and its relationship with the disease. Thirty PD patients (PD group) who treated at Xuanwu Hospital of Capital University of Medical Sciences between January 2017 and December 2019 were selected as the research objects and 30 healthy subjects were enrolled in the study during the same period as the control group. Cognitive function was assessed according to the Mini-Mental State Examination (MMSE) and Montreal Cognitive Assessment (MoCA) were used to evaluate cognitive function, Non-Motor Symptoms Scale (NMSS) was used to evaluate severity of non-motor symptoms. The relative expression of lncRNAs in plasma was measured by PCR, and the levels of neuron-specific enolase (NSE), nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF) in plasma were measured by ELISA, and the relationship with these all indexes was analyzed. The NMSS score of PD group was significantly higher than that of the control group, while the MMSE and MoCA scores were significantly lower than that of the control group ( < 0.05); The relative expression of lncRNAs , NGF and BDNF levels of PD group were significantly lower than that of the control group, and NSE level was significantly higher than that of the control group ( < 0.05); The H&Y stage and NMSS score in PD group were negatively correlated with the relative expression of lncRNAs , the levels of NGF and BDNF ( < 0.05), and positively correlated with NSE ( < 0.05); The MMSE and MoCA scores in PD group were positively correlated with the relative expression of lncRNAs , NGF, BDNF levels ( < 0.05), and negatively correlated with NSE ( < 0.05); The relative expression of lncRNAs in PD group was positively correlated with NGF, BDNF levels ( < 0.05), and negatively correlated with NSE ( < 0.05). The expression of lncRNAs in the plasma of PD patients was downregulated compared to that of healthy control subjects, and its expression level was closely related to the aggravation of non-motor symptoms, cognitive decline, and PD stage. These associations may reflect the synergism of the increase of NSE and decrease of NGF and BDNF levels, highlighting plasma lncRNA as a new candidate biomarker of PD.

探讨帕金森病(PD)患者血浆中母源表达基因3()的游离长链非编码RNA(lncRNAs)表达水平及其与疾病的关系。选取2017年1月至2019年12月在首都医科大学宣武医院治疗的30例PD患者(PD组)作为研究对象,同期纳入30例健康受试者作为对照组。根据简易精神状态检查表(MMSE)评估认知功能,采用蒙特利尔认知评估量表(MoCA)评估认知功能,采用非运动症状量表(NMSS)评估非运动症状严重程度。采用聚合酶链反应(PCR)检测血浆中lncRNAs的相对表达,采用酶联免疫吸附测定(ELISA)检测血浆中神经元特异性烯醇化酶(NSE)、神经生长因子(NGF)和脑源性神经营养因子(BDNF)水平,并分析其与这些指标的关系。PD组NMSS评分显著高于对照组,而MMSE和MoCA评分显著低于对照组(<0.05);PD组lncRNAs的相对表达、NGF和BDNF水平显著低于对照组,NSE水平显著高于对照组(<0.05);PD组H&Y分期和NMSS评分与lncRNAs的相对表达、NGF和BDNF水平呈负相关(<0.05),与NSE呈正相关(<0.05);PD组MMSE和MoCA评分与lncRNAs的相对表达、NGF、BDNF水平呈正相关(<0.05),与NSE呈负相关(<0.05);PD组lncRNAs的相对表达与NGF、BDNF水平呈正相关(<0.05),与NSE呈负相关(<0.05)。与健康对照受试者相比,PD患者血浆中lncRNAs的表达下调,其表达水平与非运动症状加重、认知功能下降及PD分期密切相关。这些关联可能反映了NSE升高与NGF和BDNF水平降低的协同作用,突出了血浆lncRNA作为PD新的候选生物标志物的作用。
