The aim of this study was to investigate the wound healing effects of the methanolic and aqueous extracts of N. Robson obtained by two different methods as well as its cytotoxicity, antioxidant activity, and selected phytochemical constituents.
Total phenolic and flavonoid contents were measured using spectrophotometry-based methods. The cytotoxic effects of the extracts on L929 mouse fibroblast cells were evaluated by and 2h-tetrazolium,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Moreover, migration and spreading of the treated fibroblast cells were assessed by cell scratch assay as an wound healing model. In addition, the chemical content of the species was determined by high pressure liquid chromatography (HPLC).
The results of the cytotoxicity assay indicated that the methanolic and aqueous extract did not have any cytotoxic effect on fibroblast cells at concentrations up to 500 μg/mL. Fibroblast migration was significantly increased by 62 μg/mL concentration of the aqueous extracts compared to the negative control. The extracts showed good antioxidant activity and 16 phytochemical compounds were detected by HPLC, with the highest amount for epicatechin.
The results showed that extracts have wound healing potential and contain several important antioxidant phenolic compounds. This species deserves further investigation aiming to isolate and identify the active compounds.
本研究旨在调查通过两种不同方法获得的罗布恩猪笼草甲醇提取物和水提取物的伤口愈合效果,以及其细胞毒性、抗氧化活性和选定的植物化学成分。
采用基于分光光度法的方法测定总酚和黄酮含量。通过2h-四唑,5-二甲基噻唑-2-基)-2,5-二苯基四氮唑溴盐法评估提取物对L929小鼠成纤维细胞的细胞毒性作用。此外,通过细胞划痕试验作为伤口愈合模型评估处理后的成纤维细胞的迁移和扩散。另外,通过高压液相色谱法(HPLC)测定该物种的化学成分。
细胞毒性试验结果表明,甲醇提取物和水提取物在浓度高达500μg/mL时对成纤维细胞没有任何细胞毒性作用。与阴性对照相比,62μg/mL浓度的水提取物显著增加了成纤维细胞的迁移。提取物表现出良好的抗氧化活性,通过HPLC检测到16种植物化学成分,表儿茶素含量最高。
结果表明,提取物具有伤口愈合潜力,并含有几种重要的抗氧化酚类化合物。该物种值得进一步研究,以分离和鉴定活性化合物。
