The emergence of carbapenem-resistant Pseudomonas aeruginosa is one of the most important challenges in a healthcare setting. The aim of this study is double-locus sequence typing (DLST) typing of bla positive P. aeruginosa isolates.
Twenty-nine bla positive isolates were collected during three years of study from different cities in Iran. Modified hodge test (MHT), double-disk synergy test (DDST) and double-disk potentiation test (DDPT) was performed for detection of carbapenemase and metallo-beta-lactamase (MBL) producing bla positive P. aeruginosa isolates. The antibiotic resistance genes were considered by PCR method. Clonal relationship of bla positive was also characterized using DLST method.
Antibiotic susceptibility pattern showed that all isolates were resistant to imipenem and ertapenem. DDST and DDPT revealed that 15/29 (51.8%) and 26 (89.7%) of bla positive isolates were MBL producing isolates, respectively. The presence of bla bla, bla and bla genes were detected in 86.2%, 41.4%, 34.5% and 3.5% isolates, respectively. DLST typing results revealed the main cluster were DLST 25-11 with 13 infected or colonized patients.
The presence of bla gene with other MBLs encoding genes in P. aeruginosa is a potential challenge in the treatment of microorganism infections. DLST showed partial diversity among 29 bla positive isolates.
耐碳青霉烯铜绿假单胞菌的出现是医疗环境中面临的最重要挑战之一。本研究的目的是对bla 阳性铜绿假单胞菌分离株进行双基因座序列分型(DLST)分型。
在三年的研究中,从伊朗不同城市收集了 29 株 bla 阳性分离株。改良 Hodge 试验(MHT)、双碟协同试验(DDST)和双碟增效试验(DDPT)用于检测产碳青霉烯酶和金属β-内酰胺酶(MBL)的 bla 阳性铜绿假单胞菌分离株。采用 PCR 法检测抗生素耐药基因。采用 DLST 法对 bla 阳性的克隆关系进行特征分析。
药敏试验结果显示,所有分离株均对亚胺培南和厄他培南耐药。DDST 和 DDPT 显示,15/29(51.8%)和 26(89.7%)bla 阳性分离株为 MBL 产生株。86.2%、41.4%、34.5%和 3.5%的分离株分别检出 bla bla、bla 和 bla 基因。DLST 分型结果显示,主要聚类为 DLST 25-11,有 13 例感染或定植患者。
铜绿假单胞菌中 bla 基因与其他 MBL 编码基因的存在是治疗微生物感染的潜在挑战。DLST 显示 29 株 bla 阳性分离株之间存在部分多样性。
