Lung cancer is the leading cause of death among cancer patients. The majority of lung cancer is the Non-Small Lung Carcinoma (NSLC). This study evaluated the potency of brazilin isolated from Caesalpinia sappan wood to induce apoptosis on non-small lung carcinoma cell line, A549, by examining the expression of p53, caspase-9, and caspase-3.
Brazilin was isolated from Caesalpinia sappan wood following a guided assay and it was determined by using Brazilin®SIGMA as standard. The activity of brazilin on the growth of A549 cell line was analysed by MTT assay and the apoptosis was evaluated by flowcytometer following Annexin V (FITC) and PI staining. The expression of p53, caspase-9, and caspase-3 was examined by immunocytochemistry.
The IC50 of brazilin on A549 cell line was 43µg/mL. Cell treatment with 20 µg/mL and 40 µg/mL of brazilin significantly increased early apoptosis (p<0.001). Cell treatment with 40 µg/mL of Brazilin significantly increased late apoptosis (p<0.001). Brazilin significantly increased the expression of p53, Caspase-9, and caspase-3 (p<0.001).
This study showed evidence of the activity of brazilin to induce intrinsic apoptosis on a NSLC cell line A549.
肺癌是癌症患者死亡的主要原因。大多数肺癌是非小细胞肺癌(NSCLC)。本研究通过检测 p53、caspase-9 和 caspase-3 的表达,评估从苏木中分离得到的巴西苏木素诱导非小细胞肺癌细胞系 A549 凋亡的能力。
按照指导检测方案从苏木中分离巴西苏木素,并使用巴西苏木素®SIGMA 作为标准进行鉴定。MTT 法分析巴西苏木素对 A549 细胞系生长的活性,用 Annexin V(FITC)和 PI 染色后通过流式细胞仪评估细胞凋亡。用免疫细胞化学法检测 p53、caspase-9 和 caspase-3 的表达。
巴西苏木素对 A549 细胞系的 IC50 为 43µg/mL。用 20µg/mL 和 40µg/mL 的巴西苏木素处理细胞可显著增加早期凋亡(p<0.001)。用 40µg/mL 的巴西苏木素处理细胞可显著增加晚期凋亡(p<0.001)。巴西苏木素显著增加了 p53、Caspase-9 和 caspase-3 的表达(p<0.001)。
本研究表明巴西苏木素具有诱导非小细胞肺癌细胞系 A549 发生内在凋亡的活性。
