Abstract

Promising advances have been made in mesenchymal stem cell transplantation to reinducethe immune tolerance in neuroinflammatory animal models and multiple sclerosis (MS)patients. The available evidence demonstrated that immunomodulatory effects of mesenchymalstem cell are particularly exerted through releasing exosomes to their environment. Wetherefore, aimed to comparatively assess the potential effect of mesenchymal stem cells andmesenchymal stem cells-derived exosomes on proliferation and function of the CD4CD25 conventional T cells, isolated from relapsing-remitting MS patients. Mesenchymal stem cells were isolated from human umbilical cord tissues and usedfor exosome isolation via ultracentrifugation. Both mesenchymal stem cells and mesenchymalstem cells-derived exosomes were evaluated for their anti-inflammatory effects againstthe proliferation of T cells isolated from two groups of individuals , MS patients andhealthy subjects. Cytokine production of conventional T cells (interferon-γ, interleukin-10, andinterleukin-17) was also assessed, using flow cytometry for the patients and healthy individuals. Here, evidence shows that MSCs and MSC-derived exosomes dampen proliferationand percentage of conventional T cells that produce IFN-γ (healthy control: < 0.001) andinterleukin-17 (healthy control: <0.001, MS patients: < 0.001), with a significant increaseof IL-10 producing cells in the patients and healthy individuals. Surprisingly, MSC-derivedexosomes demonstrated higher immune-modulating properties on conventional T cellsresponses, compared to mesenchymal stem cells (MSCs). The current study, provides a novel approach of exocytosis on autoimmune therapy.In particular, Mesenchymal stem cell -derived exosomes, which are cell-derived biologics,could be considered as an alternative for Mesenchymal stem cells in treating MS.

间充质干细胞移植在神经炎症动物模型和多发性硬化症(MS)患者中诱导免疫耐受方面取得了有前景的进展。现有证据表明,间充质干细胞的免疫调节作用尤其通过向其周围环境释放外泌体来发挥。因此,我们旨在比较评估间充质干细胞和间充质干细胞衍生的外泌体对从复发缓解型MS患者中分离出的CD4CD25常规T细胞增殖和功能的潜在影响。从人脐带组织中分离间充质干细胞,并通过超速离心用于外泌体分离。评估了间充质干细胞和间充质干细胞衍生的外泌体对从两组个体(MS患者和健康受试者)中分离出的T细胞增殖的抗炎作用。还使用流式细胞术评估了患者和健康个体中常规T细胞(干扰素-γ、白细胞介素-10和白细胞介素-17)的细胞因子产生情况。在这里,证据表明间充质干细胞和间充质干细胞衍生的外泌体抑制了产生干扰素-γ(健康对照:<0.001)和白细胞介素-17(健康对照:<0.001,MS患者:<0.001)的常规T细胞的增殖和百分比,患者和健康个体中产生白细胞介素-10的细胞显著增加。令人惊讶的是,与间充质干细胞(MSCs)相比,间充质干细胞衍生的外泌体对常规T细胞反应表现出更高的免疫调节特性。当前的研究为自身免疫治疗提供了一种新的胞吐作用方法。特别是,间充质干细胞衍生的外泌体作为细胞衍生的生物制品,在治疗MS方面可被视为间充质干细胞的替代物。
