A novel Gram-stain-positive, aerobic, non-motile and rod-shaped bacterium, designated strain NC76-1, was isolated from soil from a field that had undergone seven years continuous maize cropping from Liuba town located in Zhangye city, Gansu province, PR China. Colonies of strain NC76-1 were white, opaque and circular with a convex shape. The isolate was found to be able to grow at 10-40 °C (optimum 30 °C), pH 6.0 to 12.0 (optimum 7.0-8.0) and with 0-5.0 % (w/v) NaCl (optimum 0%). On the basis of the results of 16S rRNA gene sequence analysis, the strain fell within the clade of the genus , showing the highest sequence similarities with 40 (97.4%), CIP 108388 (97.0%), subsp. TAN 31504 (96.7%) and M1T8B10 (96.7%). The average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values between NC76-1 and its closest relatives, 40, CIP 108388, subsp. TAN 31504 and M1T8B10 were ≤73.5 % and 20.3%, respectively. The genomic DNA G+C content of NC76-1 was 61.5 mol%. It presented MK-11 as the predominant menaquinone. The major cellular fatty acids were anteiso-C (49.2 %) and iso-C (35.7%). The major polar lipids were found to be diphosphatidyglycerol, phosphatidylglycerol, phosphatidylethanolamine, aminoglycolipid, five glycolipid and one unidentified lipids. The cell wall amino acids were 2,4-diaminobutyric acid, alanine, glutamic acid, glycine and threonine. On the basis of the phylogenetic, phenotypic and chemotaxonomic characteristics, strain NC76-1 is concluded to represent a novel species within the genus , for which the name sp. nov. is proposed. The type strain is NC76-1 (GDMCC 1.2286= JCM 34651).
从中国甘肃省张掖市柳坝镇一块连续七年种植玉米的农田土壤中分离出一株新型革兰氏阳性、需氧、无运动性的杆状细菌,命名为NC76-1菌株。NC76-1菌株的菌落为白色、不透明、圆形,呈凸起状。该分离株能够在10-40°C(最适温度30°C)、pH 6.0至12.0(最适pH 7.0-8.0)和0-5.0%(w/v)NaCl(最适0%)的条件下生长。基于16S rRNA基因序列分析结果,该菌株属于 属的进化分支,与 40(97.4%)、CIP 108388(97.0%)、 亚种TAN 31504(96.7%)和M1T8B10(96.7%)的序列相似性最高。NC76-1与其最接近的亲缘种 40、CIP 108388、 亚种TAN 31504和M1T8B10之间的平均核苷酸同一性(ANI)和数字DNA-DNA杂交(dDDH)值分别≤73.5%和20.3%。NC76-1的基因组DNA G+C含量为61.5 mol%。其主要的甲基萘醌为MK-11。主要的细胞脂肪酸为anteiso-C(49.2%)和iso-C(35.7%)。主要的极性脂质为双磷脂酰甘油、磷脂酰甘油、磷脂酰乙醇胺、氨基糖脂、五种糖脂和一种未鉴定的脂质。细胞壁氨基酸为2,4-二氨基丁酸、丙氨酸、谷氨酸、甘氨酸和苏氨酸。基于系统发育、表型和化学分类学特征,得出NC76-1菌株代表 属内一个新物种的结论,为此提出新物种名称 sp. nov.。模式菌株为NC76-1(GDMCC 1.2286 = JCM 34651)。
