Abstract

Metal-organic frameworks (MOFs), also called porous coordination polymers, represent a class of crystalline porous materials made up of organic ligands and metal ions/metal clusters. Herein, an overview of the preparation of different metal-organic frameworks and the recent advances in MOF-based stimuli-responsive drug delivery systems (DDSs) with the drug release mechanisms including pH-, temperature-, ion-, magnetic-, pressure-, adenosine-triphosphate (ATP)-, HS-, redox-, responsive, and photoresponsive MOF were rarely introduced. The combination therapy containing of two or more treatments can be enhanced treatment effectiveness through overcoming limitations of monotherapy. Photothermal therapy (PTT) combined with chemotherapy (CT), chemotherapy in combination with PTT or other combinations were explained to overcome drug resistance and side effects in normal cells as well as enhancing the therapeutic response. Integrated platforms containing of photothermal/drug-delivering functions with magnetic resonance imaging (MRI) properties exhibited great advantages in cancer therapy.

金属-有机骨架(MOFs),也称为多孔配位聚合物,代表了一类由有机配体和金属离子/金属簇组成的结晶多孔材料。本文综述了不同金属-有机骨架的制备方法以及基于 MOF 的刺激响应型药物输送系统(DDS)的最新进展,包括 pH、温度、离子、磁、压力、三磷酸腺苷(ATP)、HS、氧化还原、响应和光响应 MOF 的药物释放机制。包含两种或多种治疗方法的联合治疗可以通过克服单一疗法的局限性来增强治疗效果。光热疗法(PTT)联合化学疗法(CT)、化学疗法与 PTT 或其他联合疗法的结合,旨在克服耐药性和正常细胞的副作用,并增强治疗反应。具有磁共振成像(MRI)特性的光热/药物输送功能的集成平台在癌症治疗中具有很大的优势。
