Abstract

A Gram-stain-positive, nonmotile, endospore-forming and short rod bacterium, designated as CN-4, was isolated from farm soil collected at Nanchang city, PR China. Strain CN-4 grew at 15-42 °C (optimum at 30 °C), pH 6.0-9.0 (optimum at pH 7.0) with NaCl content of 0-4% (optimum at 0.5%), and was able to fix nitrogen. The result of 16S rRNA gene sequence analysis found that strain CN-4 was most closely associated with Paenibacillus albidus Q4-3 (96.8% similarity), P. apii 7124 (96.7% similarity) and P. tianjinensis TB2019 (96.5% similarity). In the phylogenetic trees, strain CN-4 was clustered in genus Paenibacillus but formed a distinct clade. The major cellular fatty acids (≥ 5%) were anteiso-C, iso-C, C, and anteiso-C, the major polar lipids were diphosphatidylglycerol, phosphatidylglycerol, three unidentified aminophospholipids, and five unidentified lipids. The cell wall peptidoglycan was meso-DAP, DNA G + C content was 53.7%. According to the taxonomic data, strain CN-4 represents a new species within genus Paenibacillus, as Paenibacillus nanchangensis sp. nov, with strain CN-4 (= MCCC 1K09410 = KCTC 43751) as type strain.

从中国南昌采集的农田土壤中分离出一株革兰氏染色阳性、不运动、产芽孢的短杆状细菌,命名为CN-4。菌株CN-4在15-42℃(最适温度30℃)、pH 6.0-9.0(最适pH 7.0)、NaCl含量0-4%(最适0.5%)的条件下生长,且能够固氮。16S rRNA基因序列分析结果表明,菌株CN-4与白芽孢杆菌Q4-3(相似度96.8%)、芹菜芽孢杆菌7124(相似度96.7%)和天津芽孢杆菌TB2019(相似度96.5%)关系最为密切。在系统发育树中,菌株CN-4聚类在芽孢杆菌属,但形成一个独特的分支。主要细胞脂肪酸(≥5%)为anteiso-C、iso-C、C和anteiso-C,主要极性脂为二磷脂酰甘油、磷脂酰甘油、三种未鉴定的氨基磷脂和五种未鉴定的脂类。细胞壁肽聚糖为中-DAP,DNA G+C含量为53.7%。根据分类学数据,菌株CN-4代表芽孢杆菌属内的一个新物种,命名为南昌芽孢杆菌(Paenibacillus nanchangensis sp. nov),以菌株CN-4(=MCCC 1K09410=KCTC 43751)作为模式菌株。
