To establish a fingerprint analysis method of Qingfengtong capsule by HPLC.
The samples were extracted with 70% ethanol in an ultrasonic bath. The extracts were analyzed at 35 degrees C on a Diamonsil C18 column (4.6 mm x 250 mm, 5 microm) with 0.1 mol x L(-1) potassium dihydrogen phosphate water-solution as mobile phase A, and methanol as mobile phase B. The analysis followed a linear gradient elution program. Initial condition of the mobile phase was 10% B for 2 minutes; then changed to 90% B in 40 minutes. The flow rate was kept at 1.0 ml x min(-1) and the detector wavelength was 262 nm.
The main peaks in fingerprint chromatogram of Qingfengtong capsules were separated fairly well. The results of method validation meet the requirements for the fingerprints.
The established method can be used for the quality control of Qingfengtong capsules.
建立清风痛胶囊的高效液相色谱指纹图谱分析方法。
样品用70%乙醇在超声浴中提取。提取物在35℃下,于Diamonsil C18柱(4.6 mm×250 mm,5μm)上进行分析,以0.1 mol·L⁻¹磷酸二氢钾水溶液为流动相A,甲醇为流动相B。分析采用线性梯度洗脱程序。流动相初始条件为10%B,保持2分钟;然后在40分钟内变为90%B。流速保持在1.0 ml·min⁻¹,检测波长为262 nm。
清风痛胶囊指纹图谱中的主要峰分离良好。方法验证结果符合指纹图谱的要求。
所建立的方法可用于清风痛胶囊的质量控制。
