Abstract

OBJECTIVES

Some studies suggest that chronic low-grade inflammation is involved in insulin resistance in polycystic ovary syndrome (PCOS). This study assessed possible involvement of alteration in expression of two pro-inflammatory factors, tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in adipose tissues of PCOS rats in the impairment of insulin actions. Also, effects of resveratrol as an anti-inflammatory agent were investigated.

MATERIALS AND METHODS

Fifteen female rats (21 days old) were divided into three groups (n=5): I) Control, II) PCO-model-saline: served as PCOS rats and to induce PCOS, received subcutaneously testosterone enanthate 1 mg/100 g body weight subcutaneously for 35 days, III) PCO-model-resveratrol, after receiving testosterone, received resveratrol 10 mg/kg intraperitoneally for 28 days. The expression of and mRNAs in adipose tissues was determined by the qRT-PCR method.

RESULTS

The mRNA expression in the visceral adipose tissue of PCOS rats was increased in comparison to controls (<0.05). and mRNA expression in visceral and subcutaneous adipose tissues of polycystic ovarian rats was similar to controls. The expression of mRNA in subcutaneous adipose tissue and and mRNAs in the visceral adipose tissue of the PCO-model-resveratrol group were lower than PCOS rats (<0.05).

CONCLUSION

Increased expression of mRNA in the visceral adipose tissue of polycystic ovarian rats may be one cause of insulin resistance observed in them and resveratrol as an anti-inflammatory and anti-hyperglycemic agent may decrease the risk of diabetes by reduction of expression of pro-inflammatory cytokines TNF-α and IL-6 in PCOS patients.

目的

一些研究表明,慢性低度炎症与多囊卵巢综合征(PCOS)的胰岛素抵抗有关。本研究评估了多囊卵巢综合征大鼠脂肪组织中两种促炎因子肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)表达改变在胰岛素作用受损中的可能作用。此外,还研究了白藜芦醇作为抗炎剂的作用。

材料与方法

15只21日龄雌性大鼠分为三组(n = 5):I)对照组;II)PCO模型-生理盐水组:作为多囊卵巢综合征大鼠,为诱导多囊卵巢综合征,皮下注射庚酸睾酮1mg/100g体重,持续35天;III)PCO模型-白藜芦醇组,注射睾酮后,腹腔注射白藜芦醇10mg/kg,持续28天。采用qRT-PCR法测定脂肪组织中TNF-α和IL-6 mRNA的表达。

结果

与对照组相比,多囊卵巢综合征大鼠内脏脂肪组织中TNF-α mRNA表达增加(P<0.05)。多囊卵巢大鼠内脏和皮下脂肪组织中IL-6和TNF-α mRNA表达与对照组相似。PCO模型-白藜芦醇组皮下脂肪组织中IL-6 mRNA表达以及内脏脂肪组织中TNF-α和IL-6 mRNA表达均低于多囊卵巢综合征大鼠(P<0.05)。

结论

多囊卵巢大鼠内脏脂肪组织中TNF-α mRNA表达增加可能是其胰岛素抵抗的原因之一,白藜芦醇作为一种抗炎和抗高血糖药物,可能通过降低多囊卵巢综合征患者促炎细胞因子TNF-α和IL-6的表达来降低糖尿病风险。
