Strain H2R21, a novel actinobacterium, isolated from a forest soil sample collected from Heybeliada, Istanbul, Turkey, and a polyphasic approach was used for characterisation of the strain. Chemotaxonomic and morphological characterisation of strain H2R21 indicated that it belongs to the genus Nonomuraea. 16S rRNA gene sequence similarity showed that the strain is closely related to Nonomuraea purpurea 1SM4-01 (99.1%) and Nonomuraea solani CGMCC 4.7037 (98.4%). DNA-DNA relatedness values were found to be lower than 70% between the isolate and its phylogenetic neighbours N. purpurea 1SM4-01, N. solani CGMCC 4.7037 and Nonomuraea rhizophila YIM 67092. The whole cell hydrolysates of strain H2R21 were found to contain meso-diaminopimelic acid as the diagnostic diamino acid and glucose, madurose, mannose and ribose as the cell sugars. The polar lipids were identified as phosphatidylglycerol, diphosphatidylglycerol, phosphatidylmethylethanolamine, phosphatidylethanolamine, hydroxy-phosphatidylethanolamine, dihydroxy-phosphatidylethanolamine, phosphatidylinositol, glycophosphatidylinositol, two glycophospholipids and two unidentified lipids. The predominant menaquinones were identified as MK-9(H) and MK-9(H). The major fatty acids were found to be iso-C, iso-C 2OH and C 10-methyl. On the basis of DNA-DNA relatedness data and some phenotypic characteristics, it is evident that strain H2R21 can be distinguished from the closely related species in the genus Nonomuraea. Thus, it is concluded that strain H2R21 represents a novel species of the genus Nonomuraea, for which the name Nonomuraea insulae sp. nov. is proposed. The type strain is H2R21 (= DSM 102915 = CGMCC 4.7338 = KCTC 39769).
菌株H2R21是从土耳其伊斯坦布尔海贝利达采集的森林土壤样本中分离出的一种新型放线菌,采用多相分类法对该菌株进行了鉴定。菌株H2R21的化学分类和形态学鉴定表明它属于野野村菌属。16S rRNA基因序列相似性显示该菌株与紫色野野村菌1SM4-01(99.1%)和茄科野野村菌CGMCC 4.7037(98.4%)密切相关。该分离株与其系统发育邻种紫色野野村菌1SM4-01、茄科野野村菌CGMCC 4.7037和嗜根野野村菌YIM 67092之间的DNA-DNA杂交值低于70%。发现菌株H2R21的全细胞水解产物含有内消旋二氨基庚二酸作为诊断性二氨基酸,以及葡萄糖、马杜拉糖、甘露糖和核糖作为细胞糖类。极性脂质被鉴定为磷脂酰甘油、双磷脂酰甘油、磷脂酰甲基乙醇胺、磷脂酰乙醇胺、羟基磷脂酰乙醇胺、二羟基磷脂酰乙醇胺、磷脂酰肌醇、糖基磷脂酰肌醇、两种糖脂和两种未鉴定的脂质。主要的甲基萘醌被鉴定为MK-9(H)和MK-9(H)。主要脂肪酸为异-C、异-C 2OH和C 10-甲基。基于DNA-DNA杂交数据和一些表型特征,显然菌株H2R21可与野野村菌属中密切相关的物种区分开来。因此,得出结论,菌株H2R21代表野野村菌属的一个新物种,为此提出名为海岛野野村菌(Nonomuraea insulae sp. nov.)。模式菌株为H2R21(= DSM 102915 = CGMCC 4.7338 = KCTC 39769)。
