Abstract

Celastrol is a natural triterpene isolated from the Chinese plant Thunder God Vine with potent antitumor activity. However, the effect of celastrol on the growth of ovarian cancer cells and is still unclear. In this study, we found that celastrol induced cell growth inhibition, cell cycle arrest in G2/M phase and apoptosis with the increased intracellular reactive oxygen species (ROS) accumulation in ovarian cancer cells. Pretreatment with ROS scavenger N-acetyl-cysteine totally blocked the apoptosis induced by celastrol. Additionally, celastrol inhibited the growth of ovarian cancer xenografts in nude mice. Altogether, these findings suggest celastrol is a potential therapeutic agent for treating ovarian cancer.

摘要

雷公藤红素是从中国植物雷公藤中分离出的一种具有强大抗肿瘤活性的天然三萜。然而,雷公藤红素对卵巢癌细胞生长的影响仍不清楚。在本研究中,我们发现雷公藤红素可诱导卵巢癌细胞生长抑制、细胞周期阻滞于G2/M期并诱导凋亡,同时细胞内活性氧(ROS)积累增加。用ROS清除剂N-乙酰半胱氨酸预处理可完全阻断雷公藤红素诱导的凋亡。此外,雷公藤红素可抑制裸鼠体内卵巢癌异种移植物的生长。总之,这些发现表明雷公藤红素是治疗卵巢癌的一种潜在治疗剂。
