Abstract

Growth traits are the economically important traits of sheep, and screening for genes related to growth and development is helpful for the genetic improvement of ovine growth traits. The fatty acid desaturase 3 () is one of the important genes affecting the synthesis and accumulation of polyunsaturated fatty acids in animals. In this study, the expression levels of the gene and polymorphism of the gene associated with growth traits in Hu sheep were detected using quantitative real-time PCR (qRT-PCR), Sanger sequencing, and KAspar assay. The result showed that the expression levels of the gene were widely expressed in all tissues, and the expression level of in the lung was significantly higher than in other tissues ( < .05). Then, the polymorphism locus g. 2918 A > C was detected in intron 2 of the gene, and associated analysis showed that the mutation in the gene was associated significantly with growth traits (including body weight, body height, body length, and chest circumference,  < .05). Therefore, individuals with genotype showed significantly better growth traits than those with genotype, and gene could be a candidate gene for improving growth traits in Hu sheep.

生长性状是绵羊重要的经济性状,筛选与生长发育相关的基因有助于绵羊生长性状的遗传改良。脂肪酸去饱和酶3(FADS3)是影响动物多不饱和脂肪酸合成与积累的重要基因之一。本研究采用实时荧光定量PCR(qRT-PCR)、Sanger测序和竞争性等位基因特异性PCR(KASP)技术检测了湖羊中FADS3基因的表达水平及其与生长性状相关的基因多态性。结果表明,FADS3基因在所有组织中均广泛表达,且在肺中的表达水平显著高于其他组织(P<0.05)。随后,在FADS3基因内含子2中检测到多态性位点g.2918 A>C,关联分析表明该基因的突变与生长性状(包括体重、体高、体长和胸围)显著相关(P<0.05)。因此,CC基因型个体的生长性状显著优于AA基因型个体,FADS3基因可能是改善湖羊生长性状的候选基因。
