Abstract

A new condensing reagent 1-(2,4,6,-triisopropylbenzenesulfonyl)-5-(pyridin-2-yl)tetrazolide (TPSPy) was found to give one diastereoisomer of dinucleoside monophosphate aryl esters. Several oligodeoxynucleotide blocks were prepared using this reagent. A heptadecanucleotide, dTATCCCTTGCGGTGATA, which had the same sequence as the lambda cro binding DNA sequence was synthesized by condensing mono-, tri- and dodecanucleotide blocks using this reagent on a polystyrene support.

摘要

一种新型缩合试剂1-(2,4,6-三异丙基苯磺酰基)-5-(吡啶-2-基)四唑化物（TPSPy）被发现可生成二核苷单磷酸芳基酯的一种非对映异构体。使用该试剂制备了几个寡脱氧核苷酸片段。通过在聚苯乙烯载体上使用该试剂缩合单核苷酸、三核苷酸和十二核苷酸片段，合成了一种十七聚体核苷酸dTATCCCTTGCGGTGATA，其序列与λ cro结合DNA序列相同。
