To establish an HPLC method for determination of chlorogenic acid and hydrochlorothiazide in Zhenjujiangyapian.
The HPLC method was carried on C18 column using methanol-0.1% phosphoric acid(20: 80) as mobile phase, and the detection wavelength was 327 nm, the flow rate was 1.0 mL x min(-1) and the temperature of column was 40 degrees C.
In the HPLC method, the calibration curve for chlorogenic acid, hydrochlorothiazide were linear in the range of 0.049 6-0.496 (r = 0.999 5) and 1.002-10.02 microg (r = 0.999 8). The average recovery for chlorogenic acid, hydrochlorothiazide were 101.0% and 100. 1%. RSD were 2.0% and 1.4% (n = 9), respectively.
The method is convenient, precise and reliable for determining the content of chlorogenic acid and hydrochlorothiazide in Zhenjujiangyapian.
建立高效液相色谱法测定珍菊降压片中绿原酸和氢氯噻嗪的含量。
采用高效液相色谱法,以十八烷基硅烷键合硅胶为填充剂;以甲醇 - 0.1%磷酸溶液(20∶80)为流动相;检测波长为327nm;流速为1.0ml·min-1;柱温为40℃。
绿原酸在0.049 6~0.496μg范围内线性关系良好(r = 0.999 5),氢氯噻嗪在1.002~10.02μg范围内线性关系良好(r = 0.999 8)。绿原酸平均回收率为101.0%,氢氯噻嗪平均回收率为100.1%,相对标准偏差分别为2.0%和1.4%(n = 9)。
该方法简便、准确、可靠,可用于珍菊降压片中绿原酸和氢氯噻嗪的含量测定。
