To study the effects of Bushen Zhuyun Decoction (BZD) on the reproductive capacity of mice.
Totally 60 female Kunming mice were randomly divided into the experimental group and the control group. BZD was perfused by gastric tube to mice in the experimental group, twice daily. Meanwhile, mice in the control group were administered with normal saline by gastrogavage. Mice were sacrificed on the 10th day and the 20th day of medication respectively, 15 mice each time in each group. The serum levels of estradiol and progesterone were detected by radioimmunoassay. The morphological changes of uterus and ovary were observed using HE staining. The expressions of leukaemia inhibitory factor (LIF), epidermal growth factor (EGF), and calcitionin (CT) were determined using immunohistochemical assay.
Compared with the control group at the same time point, the weights of uterus and ovary increased, the serum levels of estradiol and progestogen increased, the expressions of LIF, CT, and EGF increased in the experimental group on the 10th day and the 20th day of medication (P <0.05, P <0.01). Better results were shown on the 20th day of medication (P <0.05). Histological results showed increased ovarian follicle numbers, increased endometrial gland numbers, endometrial hyperplasia (stratiform arranged), and increased stromal cells in the experimental group, especially on the 20th day of medication.
BZD could improve the reproductive capacity by advancing the development of generative organs, promoting the secretion of estradiol and progestogen and the follicular growth, and increasing the receptivity of endometrium.
研究补肾助孕汤(BZD)对小鼠生殖能力的影响。
将60只雌性昆明小鼠随机分为实验组和对照组。实验组小鼠每日经胃管灌注BZD两次。同时,对照组小鼠经胃管给予生理盐水。分别在给药第10天和第20天处死小鼠,每组每次15只。采用放射免疫法检测血清雌二醇和孕酮水平。用苏木精-伊红(HE)染色观察子宫和卵巢的形态变化。采用免疫组织化学法检测白血病抑制因子(LIF)、表皮生长因子(EGF)和降钙素(CT)的表达。
与同一时间点的对照组相比,给药第10天和第20天实验组小鼠子宫和卵巢重量增加,血清雌二醇和孕激素水平升高,LIF、CT和EGF表达增加(P<0.05,P<0.01)。给药第20天效果更明显(P<0.05)。组织学结果显示,实验组卵巢卵泡数量增加、子宫内膜腺体数量增加、子宫内膜增生(呈层状排列)以及基质细胞增多,尤其在给药第20天。
补肾助孕汤可通过促进生殖器官发育、促进雌二醇和孕激素分泌及卵泡生长、增加子宫内膜容受性来提高生殖能力。
