A lipase producer psychrophilic microorganism isolated from Arctic sample was studied. The genomic DNA of the isolate was extracted using modified CTAB method. Identification of the isolate by morphological and 16S rRNA sequence analysis revealed that the isolate is closely related to (97% similarity). was determined as positive lipase producer based on the plate screening using specific and sensitive plate assay of Rhodamine B. The PCR result using sp.'s full lipase gene sequence as the template primers emphasised a possible lipase gene at 900 bp band size. The gene is further cloned in a suitable vector system for expression of lipase.
对从北极样本中分离出的一种产脂肪酶嗜冷微生物进行了研究。采用改良的CTAB法提取该分离株的基因组DNA。通过形态学和16S rRNA序列分析对该分离株进行鉴定,结果表明该分离株与(相似度97%)密切相关。基于使用罗丹明B特异性灵敏平板检测法的平板筛选,确定该分离株为阳性脂肪酶产生菌。以该菌的全脂肪酶基因序列为模板引物的PCR结果显示,在900 bp条带大小处有一个可能的脂肪酶基因。该基因进一步克隆到合适的载体系统中用于脂肪酶的表达。
