Abstract

Metabolic syndrome includes a series of metabolic abnormalities that leads to diabetes mellitus and cardiovascular diseases. Plant extracts, due to their unique advantages like anti-inflammatory, antioxidant, and insulin sensitizing properties, are interesting therapeutic options to manage MetS; however, the poor solubility and low bioavailability of lipophilic bioactive components in the herbal extracts are two critical challenges. Nano-scale delivery systems are suitable to improve delivery of herbal extracts. This review, for the first time, focuses on nanoformulations of herbal extracts in MetS and related complications. Included studies showed that several forms of nano drug delivery systems such as nanoemulsions, solid lipid nanoparticles, nanobiocomposites, and green-synthesized silver, gold, and zinc oxide nanoparticles have been developed using herbal extracts. It was shown that the method of preparation and related parameters such as temperature and type of polymer are important factors affecting physicochemical stability and therapeutic activity of the final product. Many of these formulations could successfully decrease the lipid profile, inflammation, oxidative damage, and insulin resistance in and models of MetS-related complications. Further studies are still needed to confirm the safety and efficacy of these novel herbal formulations for clinical application.

代谢综合征包括一系列导致糖尿病和心血管疾病的代谢异常。植物提取物因其具有抗炎、抗氧化和胰岛素增敏等独特优势,是管理代谢综合征的有趣治疗选择;然而,草药提取物中亲脂性生物活性成分的低溶解度和低生物利用度是两个关键挑战。纳米级递送系统适合改善草药提取物的递送。本综述首次聚焦于代谢综合征及其相关并发症中草药提取物的纳米制剂。纳入的研究表明,已经利用草药提取物开发了几种形式的纳米药物递送系统,如纳米乳液、固体脂质纳米粒、纳米生物复合材料以及绿色合成的银、金和氧化锌纳米颗粒。结果表明,制备方法以及温度和聚合物类型等相关参数是影响最终产品物理化学稳定性和治疗活性的重要因素。这些制剂中的许多能够成功降低代谢综合征相关并发症的动物模型中的血脂水平、炎症、氧化损伤和胰岛素抵抗。仍需要进一步研究来证实这些新型草药制剂临床应用的安全性和有效性。
