Transfection is a modern and powerful method used to insert foreign nucleic acids into eukaryotic cells. The ability to modify host cells' genetic content enables the broad application of this process in studying normal cellular processes, disease molecular mechanism and gene therapeutic effect. In this review, we summarized and compared the findings from various reported literature on the characteristics, strengths, and limitations of various transfection methods, type of transfected nucleic acids, transfection controls and approaches to assess transfection efficiency. With the vast choices of approaches available, we hope that this review will help researchers, especially those new to the field, in their decision making over the transfection protocol or strategy appropriate for their experimental aims.
转染是一种用于将外源核酸导入真核细胞的现代且强大的方法。修饰宿主细胞遗传内容的能力使得该过程在研究正常细胞过程、疾病分子机制和基因治疗效果方面得到广泛应用。在本综述中,我们总结并比较了各种已报道文献中关于不同转染方法的特点、优势和局限性、转染核酸的类型、转染对照以及评估转染效率的方法等方面的研究结果。鉴于有大量的方法可供选择,我们希望本综述能帮助研究人员,尤其是该领域的新手,针对适合其实验目的的转染方案或策略做出决策。
