Taking leads from the available research, we aimed to develop a synergy-based herbal combination of (TC), (PE) and (PN) Also, evaluating their synergistic effect on CP-induced immunosuppression in mice model and exploring the possible mechanisms involved in reversing the damage.
The immunomodulatory activity of combination, of TC stem, PE fruits, and PN dried fruits, was determined by assays (splenocyte proliferation and pinocytic activity of peritoneal macrophages of mice) and study using CP-induced immunosuppression model in Swiss Albino mice. The ratio was optimized for combining three by MTT assay. The combination was further evaluated for anti-oxidant activity by DPPH scavenging method and quantified for its bioactive metabolites by HPTLC. Serum collected on day 0, 4, 7 and 14 was employed for estimation of haematogram (haematocrit, TLC, DLC, and haemoglobin, etc) and immune parameters (IL-10, IL-6 and TNF-α) by ELISA.
The study demonstrated, that combination of herbal extracts at an intermediate dose could inhibit the proliferation of spleen cells and peritoneal macrophages (P ≤ 0.0001) and induce suppression of pro-inflammatory mediators, and also certified that combination exerts synergized effects. The results showed that the combination possess potential antioxidant activity by DPPH scavenging method (IC-113.5 µg/ml). It was identified that combination significantly (P ≤ 0.0001) improved the immune markers, haematogram parameters, and histological parameters, with maximum protection offered by an intermediate dose.
The results suggested that present combination could be further explored clinically as potent synergy-based therapeutic approach for immune modulation.
借鉴现有研究成果,我们旨在开发一种基于协同作用的草药组合,包含(具体草药1名称)(TC)、(具体草药2名称)(PE)和(具体草药3名称)(PN)。此外,评估它们对环磷酰胺(CP)诱导的小鼠免疫抑制的协同作用,并探索逆转损伤所涉及的可能机制。
通过实验(小鼠脾细胞增殖和腹腔巨噬细胞的吞噬活性)和使用瑞士白化小鼠的CP诱导免疫抑制模型进行研究,确定(具体草药1名称)茎、(具体草药2名称)果实和(具体草药3名称)干果组合的免疫调节活性。通过MTT法优化三种草药组合的比例。通过DPPH清除法进一步评估该组合的抗氧化活性,并通过高效薄层色谱法对其生物活性代谢物进行定量。在第0、4、7和14天收集的血清用于通过ELISA评估血常规(血细胞比容、白细胞总数、白细胞分类计数和血红蛋白等)和免疫参数(IL - 10、IL - 6和TNF - α)。
研究表明,中等剂量的草药提取物组合可抑制脾细胞和腹腔巨噬细胞的增殖(P≤0.0001),并诱导促炎介质的抑制,还证明该组合具有协同作用。结果表明,通过DPPH清除法该组合具有潜在的抗氧化活性(IC - 113.5μg/ml)。确定该组合显著(P≤0.0001)改善了免疫标志物、血常规参数和组织学参数,中等剂量提供了最大保护。
结果表明,目前的组合可作为一种基于协同作用的有效免疫调节治疗方法进一步进行临床探索。
